Summary: Following human cytomegalovirus (HCMV) infection, the aryl hydrocarbon receptor (AhR) is activated and alters the levels of numerous RNAs, including those related to cell cycle progression. AhR knockdown alleviates the G1/S cell cycle block in HCMV-infected fibroblasts, providing mechanistic insight into the virus-host cell interaction.

Summary: Carvacrol, a TRPM7 inhibitor, was found to have anti-cancer potentials against breast cancer cells by regulating cell cycle and apoptosis. Summary: R-loops are three-stranded structures formed by annealing of nascent transcripts and template DNA, playing important roles in physiological processes. Dysregulation of R-loop metabolism may pose a threat to genome stability. Depletion of RAD51 and inhibition by B02 did not prevent R-loop formation in human cells, while B02 treatment may lead to premature origin firing caused by RAD51-dependent accumulation of R-loops.

Summary: This study identified ATR-CHK1 signaling as a therapeutic target for liver cancer. Pharmacological inhibition of ATR or CHK1 leads to proliferation inhibition in liver cancer cells with high replication stress levels. CDC7 inhibitors induce DNA replication stress in cells resistant to ATR or CHK1 inhibition, showing strong synergy with ATR or CHK1 inhibitors.

Summary: Cytomegaloviruses induce significant changes in cell cycle regulation to enable replication of their DNA genomes. They establish a unique cell cycle arrest at the G(1)/S transition, while activating S phase-specific gene expression and nucleotide metabolism. Despite similarities in cell cycle regulation between human and murine CMVs, there are considerable differences in viral cell cycle effectors and mechanisms of action.

The study revealed how perturbations of critical host and viral factors can alter the progression and timing of infection.
