Summary: IF1 is a natural inhibitor protein that blocks the catalysis and rotation of mitochondrial FoF1 ATP synthase. It inhibits ATP hydrolysis by F-1, but this inhibition is relieved under ATP synthesis conditions. The study shows that IF1-inhibited F-1 is activated only when F-1 rotates in the clockwise direction, explaining the condition-dependent mechanism of IF1 inhibition. Investigation of mutant IF1 with N-terminal truncations reveals the importance of its interaction with the gamma subunit and the role of the middle long helix in the inhibition of F-1.

Summary: Adrenal medullary chromaffin (AMC) cells in the perinatal period and carotid body glomus cells after birth respond to hypoxia with catecholamine secretion. The hypoxia detection mechanism in such O2-sensitive cells is still not well defined. One hypothesis is that a decrease in cellular ATP may be involved in the hypoxia detection.

Summary: IF1 is an endogenous inhibitor of ATP synthase, involved in mitochondrial functions, energy metabolism, redox balance, and cell fate. Despite recent paradigm shifts regarding IF1 functions, its study in entire organisms and different organs remains limited. The investigation of IF1 at multiple levels and in various pathophysiological conditions may help uncover therapeutic strategies in human pathology.

Summary: This study analyzed the association between plasma ATPase inhibitory factor 1 (IF1) levels and markers of glucose homeostasis as well as the development of new-onset diabetes (NOD) in individuals with prediabetes. The results showed that IF1 levels were negatively correlated with clinical variables associated with obesity and insulin resistance, and positively correlated with markers of cardiometabolic health. Furthermore, the study found a significant association between IF1 levels and the risk of type 2 diabetes mellitus (T2DM).

Summary: F-ATP synthase inhibitory factor 1 (IF1) is an intrinsic inhibitor of F-ATP synthase. This study revealed that IF1 interacts with phosphorylated c-Myc and PGC1a, regulating their activity and promoting glycolysis. Additionally, IF1 inhibits oxidative respiration by interacting with PGC1a. IF1 is also required for the protective effect of hypoxia on c-Myc- and PGC1a-induced cell death, suggesting its role in regulating metabolic reprogramming and cell fate.
