Chlamydia trachomatis is a human bacterial pathogen that multiplies only within an intracellular membrane-bound vacuole, the inclusion. C. trachomatis includes ocular and urogenital strains, usually causing infections restricted to epithelial cells of the conjunctiva and genital mucosa, respectively, and lymphogranuloma venereum (LGV) strains, which can infect macrophages and spread into lymph nodes. However, C. trachomatis genomes display >98% identity at the DNA level. In this work, we studied whether C. trachomatis Inc proteins, which have a bilobed hydrophobic domain that may mediate their insertion in the inclusion membrane, could be a factor determining these different types of infection and tropisms. Analyses of polymorphisms and phylogeny of 48 Inc proteins from 51 strains encompassing the three disease groups showed significant amino acid differences that were mainly due to variations between Inc proteins from LGV and ocular or urogenital isolates. Studies of the evolutionary dynamics of inc genes suggested that 10 of them are likely under positive selection and indicated that most nonsilent mutations are LGV specific. Additionally, real-time quantitative PCR analyses in prototype and clinical strains covering the three disease groups identified three inc genes with LGV-specific expression. We determined the transcriptional start sites of these genes and found LGV-specific nucleotides within their promoters. Thus, subtle variations in the amino acids of a subset of Inc proteins and in the expression of inc genes may contribute to the unique tropism and invasiveness of C. trachomatis LGV strains.

Molecular Capture of Mycobacterium tuberculosis Genomes Directly from Clinical Samples: A Potential Backup Approach for Epidemiological and Drug Susceptibility Inferences

Summary: This study aimed to prove the feasibility of applying whole genome sequencing of Mycobacterium tuberculosis directly on clinical samples, in order to avoid the time-consuming culture isolation and the potential prolonged suboptimal antibiotic treatment. The results demonstrated that capturing and sequencing the M. tuberculosis genome directly from positive sputum samples can provide a solution for epidemiological and drug susceptibility predictions.

The Chlamydia trachomatis IncM Protein Interferes with Host Cell Cytokinesis, Centrosome Positioning, and Golgi Distribution and Contributes to the Stability of the Pathogen-Containing Vacuole

Summary: Chlamydia trachomatis is a bacterial pathogen that infects the eyes and genitourinary system in humans. Its ability to grow inside host cells in a membrane-bound compartment called an inclusion relies on effector proteins that are transported by a type III secretion system. One of these effector proteins, IncM, was found to play a role in inhibiting host cell cytokinesis and inducing multinucleation in infected cells.

Comparative complete scheme and booster effectiveness of COVID-19 vaccines in preventing SARS-CoV-2 infections with SARS-CoV-2 Omicron (BA.1) and Delta (B.1.617.2) variants: A case-case study based on electronic health records

Summary: This study aimed to estimate the comparative vaccine effectiveness between the Omicron and Delta variants in terms of primary and booster vaccination exposure. The results showed that the odds of vaccination were higher in cases infected with the Omicron variant compared to the Delta variant, but the vaccine effectiveness was reduced. The vaccine effectiveness against Omicron for complete primary vaccination and booster dose was -6.0% and 62.7%, respectively.

Summary: The multicountry monkeypox outbreak in 2022, alongside the COVID-19 pandemic, emphasized the importance of genomic surveillance and rapid pathogen whole-genome sequencing. Highly multiplexed amplicon-based sequencing provides a cost-effective and flexible approach for pathogen whole-genome sequencing, allowing for rapid response to emerging pathogens.

Chlamydial and Gonococcal Genital Infections: A Narrative Review

Rafaela Rodrigues, Pedro Vieira-Baptista, Carlos Catalao, Maria Jose Borrego, Carlos Sousa, Nuno Vale

Summary: Sexually transmitted infections (STIs) are a leading global health concern, causing significant burden in terms of disease, mortality, healthcare costs, and social stigma. Chlamydial and gonococcal infections, caused by Chlamydia trachomatis (CT) and Neisseria gonorrhoeae (NG) respectively, are the most common bacterial STIs. These infections are often asymptomatic, but can lead to severe long-term complications and increased risk of other STIs. Dual-testing for both pathogens is recommended due to similar symptoms and frequent co-occurrence. This review provides insights into the clinical aspects, diagnostic tests, treatment, and strategies for controlling CT and NG infections worldwide. Antibiotics are the primary treatment, but the increasing incidence and antibiotic resistance pose challenges, particularly for NG infections. The potential association between CT infections and tumorigenesis is also discussed, along with the molecular basis that may aid in developing effective drugs and vaccines.

Comparative Effectiveness of COVID-19 Vaccines in Preventing Infections and Disease Progression from SARS-CoV-2 Omicron BA.5 and BA.2, Portugal

Irina Kislaya, Pedro Casaca, Vitor Borges, Carlos Sousa, Bibiana I. Ferreira, Ana Fonte, Eugenia Fernandes, Carlos Matias Dias, Silvia Duarte, Jose Pedro Almeida, Ines Grenho, Luis Coelho, Rita Ferreira, Patricia Pita Ferreira, Claudia Medeiros Borges, Joana Isidro, Miguel Pinto, Luis Menezes, Daniel Sobral, Alexandra Nunes, Daniela Santos, Antonio Maia Goncalves, Luis Vieira, Joao Paulo Gomes, Pedro Pinto Leite, Baltazar Nunes, Ausenda Machado, Andre Peralta-Santos

Summary: We evaluated the effectiveness of primary and booster vaccination against SARS-CoV-2 Omicron BA.5 and BA.2 lineages in preventing infection and disease progression. Through a case-case and cohort study, we determined the odds ratios for vaccination using logistic regression and the vaccine effectiveness against disease progression through penalized logistic regression. Our findings showed that there were no reduced vaccine effectiveness for both primary and booster vaccinations against BA.5 infection. However, booster vaccine effectiveness against hospitalization was lower for BA.5 cases compared to BA.2 cases. Despite this, booster vaccination still provided substantial protection against severe disease progression from BA.5 infection.
