The presence of bacteria species in semen and sperm quality

To verify the prevalence of semen bacterial contamination and whether the contamination could decrease sperm quality. Spermiogram, semen culture, and sperm transmission electron microscopy (TEM) analysis were performed. TEM data were elaborated using a mathematical formula that calculates a fertility index (FI)-able to define patients as fertile or infertile-and the percentage of sperm apoptosis, immaturity and necrosis. We aligned the amino acid sequence of beta-tubulin with protein of the most frequent species isolated from semen. Patients were divided according to the contaminating species; in each group, we observed fertile individuals, in whom the semen quality was similar to that of controls and infertile men whose sperm quality was significantly decreased, in terms of motility, FI, apoptosis and necrosis. Partial homology between beta-tubulin and bacterial proteins was observed. Sperm bacterial contamination is quite frequent and could contribute to the deterioration of the sperm quality of infertile men.

Fatty Acid Oxidation and Pro-Resolving Lipid Mediators Are Related to Male Infertility

Summary: Specialized pro-resolving lipid mediators, such as resolvin D1 (RvD1), play a role in regulating acute inflammation and resolving it. This study investigated the implication of seminal RvD1 in human infertility and found that higher levels of RvD1 were associated with increased inflammation markers and reduced sperm quality in infertile patients. The study suggests using a panel of inflammatory markers and lipid mediators for diagnosing and treating inflammatory conditions.

Expression and immunolocalisation of PPARγ in reproductive tissues of rabbits fed n-3 PUFA-enriched diets

Summary: This study investigated the expression and localization of PPAR gamma, as well as susceptibility to lipoperoxidation, in the reproductive tissues of rabbits fed n-3 polyunsaturated fatty acid (PUFA)-enriched diets. The results showed that PPAR gamma expression was down-regulated in control rabbit testes but up-regulated in the epididymis of rabbits fed n-3 PUFAs diets. The study suggests that n-3 PUFAs diets may influence PPAR gamma activity and play a role in supporting sperm maturation.

A Dynamic Model for Estimating the Interaction of ROS-PUFA-Antioxidants in Rabbit

Seminal Levels of Omentin-1/ITLN1 in Inflammatory Conditions Related to Male Infertility and Localization in Spermatozoa and Tissues of Male Reproductive System

Summary: This study explored the relationships among omentin-1/ITLN1, semen parameters, and oxidative stress markers in different patient groups. The levels of omentin-1/ITLN1 were found to be correlated with F-2-IsoP levels and negatively correlated with sperm parameters. Infertile patients showed lower levels of omentin-1/ITLN1, while those with varicocele and genitourinary infections had higher levels. Omentin-1/ITLN1 was localized differently in sperm and male reproductive system tissues.

Effects and Mechanisms Activated by Treatment with Cationic, Anionic and Zwitterionic Liposomes on an In Vitro Model of Porcine Pre-Pubertal Sertoli Cells

Summary: This study aimed to investigate the effects of different types of liposomes on porcine prepubertal Sertoli cells (SCs) and to find a suitable carrier for in vitro testing of drugs. The results showed that anionic and cationic liposomes altered the distribution of actin and vimentin filaments and increased the phosphorylation levels of AMPK alpha. Zwitterionic liposomes, on the other hand, did not cause any changes in the cell cytoplasm. These preliminary data suggest that zwitterionic liposomes are the most suitable carrier for in vitro studies of SCs to understand the effects of molecules or drugs with potential clinical applications in the treatment of certain forms of male infertility.

Follicular Fluid Components in Reduced Ovarian Reserve, Endometriosis, and Idiopathic Infertility

Summary: This study investigated the changes and relationships of various molecules in the follicular fluid (FF) of infertile women with different reproductive conditions. It found that some biochemical components, hormones, antioxidant enzymes, F-2-Isoprostanes (F-2-IsoPs), and resolvin (Rv) D1 in the FF are associated with follicular growth, oocyte maturation, and female infertility. These findings highlight the importance of personalized therapeutic approaches and optimized assisted reproductive techniques (ART) outcomes.

Human Sperm as an In Vitro Model to Assess the Efficacy of Antioxidant Supplements during Sperm Handling: A Narrative Review

Summary: Spermatozoa produce reactive oxygen species (ROS) due to aerobic metabolism, which can have both beneficial and detrimental effects on cell function. Assisted reproductive procedures, such as cryopreservation, can lead to excessive ROS production and oxidative damage to spermatozoa. This review focuses on using human sperm as an in vitro model to study the supplementation of antioxidants in media. The presence and synergic effects of different antioxidant molecules may lead to more effective products in vitro and potentially in vivo.

Can Dietary n-3 Polyunsaturated Fatty Acids Affect Apelin and Resolvin in Testis and Sperm of Male Rabbits?

Summary: This study investigated the association of apelin and resolvin (RvD1) in the testis and sperm with the oxidative status of semen and serum testosterone in rabbits fed different diets enriched with flaxseed or fish oil. The results showed that flaxseed diet increased the levels of apelin in the testis and enhanced serum testosterone. Both enriched diets showed higher levels of malondialdehyde and RvD1 in the testis. Apelin and its receptor were also detected in ejaculated sperm.

F4-Neuroprostane Effects on Human Sperm

Summary: After incubation with F-4-NeuroPs, selected human sperm showed increased motility, membrane mitochondrial potential, and reacted acrosome percentage. Chromatin integrity did not differ between treated and control samples. PSA staining highlighted a significantly higher number of sperm with reacted acrosomes in the treated group after 4 hours. Immunofluorescence analysis revealed a dense distribution of ryanodine receptors in the midpiece and postacrosomal sheath of sperm after 2 hours, followed by a dotted distribution after 4 hours. F-4-NeuroPs at an appropriate concentration can induce sperm capacitation via ion channels, which may enhance the success of assisted fertilization.
