The development and maintenance of the sealing of the root canal system is the key to the success of root canal treatment. The resin-based adhesive material has the potential to reduce the microleakage of the root canal because of its adhesive properties and penetration into dentinal walls. Moreover, the irrigation protocols may have an influence on the adhesiveness of resin-based sealers to root dentin. Objective: The objective of the present study was to evaluate the effect of different irrigant protocols on coronal bacterial microleakage of gutta-percha/AH Plus and Resilon/Real Seal Self-Etch systems. Material and Methods: One hundred ninety pre-molars were used. The teeth were divided into 18 experimental groups according to the irrigation protocols and filling materials used. The protocols used were: distilled water; sodium hypochlorite (NaOCl)+EDTA; NaOCl+H3PO4; NaOCl+EDTA+chlorhexidine (CHX); NaOCl+H3PO4+CHX; CHX+EDTA; CHX+H3PO4; CHX+EDTA+CHX and CHX+H3PO4+CHX. Gutta-percha/AH Plus or Resilon/Real Seal SE were used as root-filling materials. The coronal microleakage was evaluated for 90 days against Enterococcus faecalis. Data were statistically analyzed using Kaplan-Meier survival test, Kruskal-Wallis and Mann-Whitney tests. Results: No significant difference was verified in the groups using chlorhexidine or sodium hypochlorite during the chemo-mechanical preparation followed by EDTA or phosphoric acid for smear layer removal. The same results were found for filling materials. However, the statistical analyses revealed that a final flush with 2% chlorhexidine reduced significantly the coronal microleakage. Conclusion: A final flush with 2% chlorhexidine after smear layer removal reduces coronal microleakage of teeth filled with gutta-percha/AH Plus or Resilon/Real Seal SE.

Investigation of microbial profile, levels of endotoxin and lipoteichoic acid in teeth with symptomatic irreversible pulpitis: a clinical study

R. Arruda-Vasconcelos, L. M. Louzada, M. Feres, P. L. Tomson, P. R. Cooper, B. P. F. A. Gomes

Summary: The microbial profile, and levels of endotoxin (LPS) and lipoteichoic acid (LTA), in infected dentine and root canals of teeth with irreversible pulpitis were investigated. Root canal preparation effectively reduced the levels of bacteria, LPS and LTA, while intracanal medication did not significantly reduce bacterial levels but led to a significant decrease in LTA levels.

Summary: The study evaluated Enterococcus faecalis strains isolated from endodontic failures for the presence of virulence genes, biofilm formation capacity, and activity of gelatinase and beta-lactamase. Results showed diverse patterns of virulence gene detection among the strains, with most capable of forming biofilm. Some strains exhibited gelatinase production, while beta-lactamase resistance was detected in different isolates.

Heating stability, physical and chemical analysis of calcium silicate-based endodontic sealers

T. B. M. Antunes, A. C. P. Janini, L. E. Pelepenko, G. F. Abuna, E. M. Paiva, M. A. C. Sinhoreti, I. M. Raimundo, B. P. F. A. Gomes, A. de-Jesus-Soares, M. A. Marciano

Summary: The study evaluated modifications in root canal sealers under different heating conditions using vibrational spectroscopy and chemical analysis. Heating significantly changed the flowability and solubility of calcium silicate-based sealers, with AH Plus being the only stable sealer after heating. Calcium silicate-based sealers exhibited high solubility, raising concerns for their clinical use.

Analysis of microorganisms in periapical lesions: A systematic review and meta-analysis

Juliana D. Bronzato, Rafael A. Bomfim, Giovanna Z. P. Hayasida, Mariana Curi, Carlos Estrela, Bruce J. Paster, Brenda P. F. A. Gomes

Summary: The study systematically reviewed the literature on the prevalence of microorganisms in endodontic periapical lesions and found that the prevalence was high, with 87% of lesions containing microorganisms. The prevalence of viable/active microorganisms was slightly lower at 82%. Different geographic areas showed statistical differences, as did viable bacteria compared to active viruses. The most common detection method for microorganisms was molecular, with Actinomyces, Fusobacterium, and Prevotella being the most prevalent bacteria.

Summary: This study evaluated the presence of Fusobacterium nucleatum in primary and secondary endodontic infections in a Brazilian population using both culture and nested PCR methods. The results showed a higher detection rate of F. nucleatum in primary infections compared to secondary ones, with nested PCR method detecting a higher prevalence. The presence of F. nucleatum was associated with various clinical features in root canals.

Towards an enhanced nanocarbon crystallization from lignin

Douglas Rocha, Jessica Luzardo, Daniele Aguiar, Sanair Massafra, Rogerio Valaski, Alexander Silva, Erlon Ferreira, Renata Simao, Braulio Archanjo, Alexei Kuznetsov, Joyce Araujo

Summary: A new approach to synthesize lignin-derived nanocarbon crystals based on lignin depolymerization is proposed in this study. The chemical structure of lignin was modified using a controlled routine involving UV light irradiation, hydrothermal carbonization, vacuum degassing, pyrolysis, and mechanical exfoliation. This method resulted in a lignin-based nanocarbon with a nanoporous framework structure and oxygenated functional groups. The catalyst-free route used in this study showed high nanocarbon yields and is an eco-friendly strategy for synthesizing low-cost carbon materials. The characterization of the lignin-derived nanocarbon included X-ray diffraction, infrared, Raman, and X-ray photoelectron spectroscopies, as well as scanning and transmission electron microscopy. The developed transformation method has the potential to provide large amounts of carbon-based materials for industrial applications, and it can be adapted to various biomass feedstocks with suitable aromatic groups for high yield polycondensation. The achieved nano-carbon yield ranged from 50 wt% to 70 wt% when starting from a commercial purified lignin.
