Hypertension is one of the most important public health problems worldwide. If undiagnosed or untreated, this pathology represents a systemic risk factor and offers unfavorable conditions for dental treatments, especially those requiring bone healing. Objectives: The purpose of this study was to demonstrate, by analysis of bone mineral density (BMD), that the alveolar bone healing process is altered in spontaneously hypertensive rats (SHRs). Material and Methods: Wistar rats and SHRs were submitted to extraction of the upper right incisor and were euthanized 7, 14, 21, 28 and 42 days after surgery. Right maxillae were collected, radiographed and analyzed using Digora software. BMD was expressed as minimum (min), middle (med) and maximum (max) in the medium (MT) and apical (AT) thirds of the dental alveolus. Results: The results were compared across days and groups. Wistar showed difference in med and max BMD in the MT between 7 and 28 and also between 14 and 28 days. The AT exhibited significant difference in med and min BMD between 7 and 28 days, as well as difference in min BMD between 28 and 42 days. SHRs showed lower lined BMD in the MT at 28 days when compared to 21 and 42 days. Differences were observed across groups in med and min BMD at day 28 in the MT and AT; and in max BMD at 14, 21 and 42 days in the MT. Conclusions: These results suggest that the alveolar bone healing process is delayed in SHRs comparing with Wistar rats.

Summary: The study aimed to evaluate the effects of vitamin D alone or in combination with teriparatide on peri-implant bone tissue in orchiectomized rats. Through an in vivo study, it was found that treatment with vitamin D and teriparatide together can increase the volume of bone tissue around the implant and improve bone quality. However, given the increasing population with bone metabolism disorders seeking dental implant rehabilitation treatment, the verification of medications that promote an increase in bone density is essential.

Normoglycemia partially recovers the disrupted osteoblast differentiation of mesenchymal stem cells induced by type 1 but not type 2 diabetes mellitus

Summary: Type 1 (T1DM) and type 2 (T2DM) diabetes mellitus affect glucose metabolism and cause bone damage through various mechanisms, including the impact on osteoblasts. This study examined the osteoblast differentiation of mesenchymal stem cells (MSCs) from rats with T1DM or T2DM and the effects of removing hyperglycemia on the osteogenic potential of these cells. The results showed that both T1DM and T2DM inhibited the osteoblast differentiation of MSCs cultured in hyperglycemic media, with T1DM having a more significant effect. Restoring normoglycemic conditions partially recovered the osteogenic potential of MSCs from rats with T1DM but not with T2DM. These findings highlight the need for targeted therapies for T1DM- or T2DM-induced bone loss, as both conditions disrupt osteoblast differentiation at different levels and likely through different mechanisms.

DNA hydrogels for bone regeneration

Dimitra Athanasiadou, Nadeen Meshry, Naara G. Monteiro, Ana C. Ervolino-Silva, Ryan Lee Chan, Christopher A. McCulloch, Roberta Okamoto, Karina M. M. Carneiro

Summary: DNA-based biomaterials have potential as biodegradable scaffolds for bone repair due to their ability to bind Ca2+ and promote HAP growth. This study describes the design and synthesis of DNA hydrogels, their interactions with osteogenic cell lines, and their promotion of new bone formation in rat calvarial wounds.

Polydioxanone Membrane Compared with Collagen Membrane for Bone Regeneration

Lilian Caldas Quirino, Pedro Henrique de Azambuja Carvalho, Renato Torres Augusto Neto, Cassio Amaro Comachio, Naara Gabriela Monteiro, Ana Claudia Ervolino-Silva, Roberta Okamoto, Valfrido Antonio Pereira-Filho

Summary: This study evaluated the bone repair in critical defects using a polydioxanone membrane (Plenum((R)) Guide) and a collagen-based membrane (Bio-Gide((R))) in rats. The results showed that the Plenum((R)) Guide group had better bone regeneration and gene expression, but there was no significant difference between the two groups in histometric and micro-CT analyses. Therefore, the absorbable polydioxanone membrane is an effective guided bone regeneration membrane.

The Local Release of Teriparatide Incorporated in 45S5 Bioglass Promotes a Beneficial Effect on Osteogenic Cells and Bone Repair in Calvarial Defects in Ovariectomized Rats

Juliani Caroline Ribeiro de Araujo, Leonardo Alvares Sobral Silva, Vinicius Almeida de Barros Lima, Tiago Moreira Bastos Campos, Paulo Noronha Lisboa Filho, Roberta Okamoto, Luana Marotta Reis de Vasconcellos

Summary: With increasing life expectancy, the prevalence of osteoporosis has risen, leading to a search for strategies to regenerate bone tissue. The sonochemical technique was used to functionalize bioglass, resulting in modified surface characteristics. In vitro tests showed favorable cell adhesion and differentiation, with no cytotoxicity observed. Animals treated with teriparatide demonstrated new bone formation, indicating positive effects on bone tissue. Further studies are required to investigate the long-term effects of the interaction between bioglass and teriparatide.

Miron, Roberta Okamoto, Michel Reis Messora

Summary: After tooth loss, bone remodeling can cause significant decreases in bone height and thickness, which can compromise the success of dental implants. Leukocyte-platelet-rich fibrin (L-PRF) has shown potential in promoting tissue regeneration and several protocols have been proposed to improve its regenerative potential. In this study, different centrifugation protocols were evaluated for their effectiveness in bone healing in rat calvaria defects. The results showed that platelet aggregates, especially with horizontal centrifugation, improve bone neoformation and have more biological potential for bone healing.

The Interplay of Raloxifene and Sonochemical Bio-Oss in Early Maxillary Sinus Bone Regeneration: A Histological and Immunohistochemical Analysis in Rabbits

Summary: The study aimed to assess the efficacy of using Raloxifene with ultrasonic processing to enhance Bio-Oss & REG; for maxillary sinus bone height reconstruction. Results indicated that while sonicated Bio-Oss & REG; performed comparably to Bio-Oss & REG; alone, the addition of Raloxifene led to an unexpected delay in bone repair. These findings stress the importance of histological evaluation for accurate bone repair assessment.

Paula Buzo Frigerio, Pedro Henrique Silva Gomes-Ferreira, Fabio Roberto De Souza Batista Juliana Moura, Idelmo Rangel Garcia Junior, Daniele Botticelli, Paulo Noronha Lisboa-Filho, Roberta Okamoto

Summary: This study evaluated the behavior of repaired bone in rats after extraction, focusing on the use of PTH (teriparatide) and Biogran (R) (a biomaterial). The results showed that the use of Biogran (R) with PTH improved bone quality in healthy rats, but did not have significant effects in orchiectomized rats. Further research is needed to determine more effective doses for postoperative bone formation.
