Analysis of the slow germination of multiple individual superdormant Bacillus subtilis spores using multifocus Raman microspectroscopy and differential interference contrast microscopy

Aim: To analyse the dynamic germination of hundreds of individual superdormant (SD) Bacillus subtilis spores. Methods and Results: Germination of hundreds of individual SD B. subtilis spores with various germinants and under different conditions was followed by multifocus Raman microspectroscopy and differential interference contrast microscopy for 12 h and with temporal resolutions of <= 30 s. SD spores germinated poorly with the nutrient germinant used to isolate them and with alternate germinants targeting the germinant receptor (GR) used originally. The mean times following mixing of spores and nutrient germinants to initiate and complete fast release of Ca-dipicolinic acid (CaDPA) (T-lag and T-release times, respectively) of SD spores were much longer than those of dormant spores. However, the Delta T-release times (T-release-T-lag) of SD spores were essentially identical to those of dormant spores. SD spores germinated almost as well as dormant spores with nutrient germinants targeting GRs different from the one used to isolate the SD spores and with CaDPA that does not trigger spore germination via GRs. Conclusions: Since (i) Delta T-release times were essentially identical in GR-dependent germination of SD and dormant spores; (ii) rates of GR-independent germination of SD and dormant spores were identical; (iii) large increases in T-lag times were the major difference in the GR-dependent germination of SD as compared with spores; and (iv) higher GR levels are correlated with shorter T-lag times, these results are consistent with the hypothesis that low levels of a GR are the major reason that some spores in a population are SD with germinants targeting this same GR. Significance and Impact of the Study: This study provides information on the dynamic germination of individual SD spores and improves the understanding of spore superdormancy.

Summary: Efforts have been made to parallelize Raman excitation and acquisition to overcome the low efficiency of conventional confocal Raman spectroscopy. A method under a compressive sensing framework has been developed to demultiplex the superimposed spectra of multiple cells during their dynamic processes, taking into account the sparse structure of cell spectra acquired at different times. Collaborative optimization of the inversion problem by penalizing sparsity from all time intervals has shown promising results, with potential applications in monitoring living cells scattering on a coverslip and improving throughput significantly.

Summary: Three new species, Novosphingobium album, Novosphingobium organovorum and Novosphingobium mangrovi, were isolated from mangrove sediments and showed potential for bioremediation. Genomic analysis identified genes related to pesticide degradation, heavy-metal resistance, nitrogen fixation, antibiotic resistance and sulfur metabolism in these isolates, indicating their biotechnological potential.

Summary: Strain 4503(T) is a newly identified species of the genus Streptomyces, isolated from mangrove sediments, with the ability to produce clinically relevant compounds. Summary: This study investigates the effects of UV radiation on Nosema bombycis spores at the single-cell level. The results demonstrate that UV radiation causes the leakage of trehalose from individual spores without significant damage to intracellular biomacromolecules. Low-dose radiation promotes spore germination, while high-dose radiation decreases germination rate. These findings suggest that UV radiation disrupts the permeability of the inner membrane and alters the spore wall, affecting spore sensing and germination ability.

Summary: In this study, single-cell Raman spectroscopy combined with MCR-ALS was applied to rapidly obtain information on the variations in the spectral intensity of major biomacromolecules (i.e., phospholipids, proteins, polysaccharides, and triglycerides) in yeast cells during ethanol fermentation at the single-cell level without any a priori information. The results revealed that lipids and cell heterogeneity play an imperative role in the strain's fermentation performance and efficiency.

Analysis of Aging Mechanism of Polystyrene Microspheres by Using Single Particle Raman Spectroscopy

Summary: This study investigated the aging mechanism of micron-sized polystyrene (PS) particles at a single-particle level by simulating UV radiation. The spectral characteristics and chemical structure changes of PS particles under UV exposure were characterized using single-particle Raman spectroscopy and scanning electron microscopy. The study found that UV radiation damages the structures of PS polymers and molecules, with a greater extent of degradation observed in smaller particle sizes.
