Multiplex PCR for detection of acquired plasmid-borne fosfomycin resistance fos genes in Escherichia coli

Summary: A rapid and reliable multiplex PCR was developed to simultaneously detect known plasmid-mediated fos genes conferring acquired resistance to fosfomycin. Our technique was tested on a collection of Escherichia coli isolates previously identified as bearing fosA, fosC, and fosL-like genes, demonstrating a sensitivity and specificity of 100%.

Progressive in vivo development of resistance to cefiderocol in Pseudomonas aeruginosa

Summary: This study reports the development of cefiderocol resistance among sequential Pseudomonas aeruginosa clinical isolates, without exposure to the drug. The resistance was driven by multifactorial mechanisms, including mutational changes in iron transporter proteins associated with drug uptake and overexpression of certain genes.

In vitro activity of cefepime/zidebactam and cefepime/taniborbactam against aztreonam/avibactam-resistant NDM-like-producing Escherichia coli clinical isolates

Summary: This study evaluated the in vitro activity of cefepime/taniborbactam and cefepime/zidebactam against aztreonam/avibactam-resistant E. coli strains. The results showed that cefepime/taniborbactam had a resistance rate of 100% against these strains, while cefepime/zidebactam was effective in all cases. This suggests that cefepime/zidebactam combination could be a promising therapeutic option.

A Selective Culture Medium for Screening Cefiderocol Resistance in Enterobacterales, Pseudomonas aeruginosa, and Acinetobacter baumannii

Summary: Cefiderocol (FDC) is an effective siderophore cephalosporin against multidrug-resistant Gram-negative bacteria. The SuperFDC medium was developed to rapidly and accurately identify FDC-resistant pathogens, which is crucial for controlling their spread. The medium showed high sensitivity and specificity in detecting FDC-resistant isolates with various resistance mechanisms.

In vitro-obtained meropenem-vaborbactam resistance mechanisms among clinical Klebsiella pneumoniae carbapenemase-producing K. pneumoniae isolates

Summary: A study was conducted to select MEM-VAB resistant mutants and decipher the resistance mechanisms. The mutants showed reduced susceptibility to various antibiotics and had mutations in the ompK36 gene. Complementation with the wildtype ompK36 gene reduced the minimal inhibitory concentrations. Additionally, the bla KPC gene copy numbers were significantly increased in the resistant mutants.

Assessment of QuantuMDx Q-POC Assay for Rapid Detection of SARS-CoV-2 Using Middle Turbinate Swabs

Summary: In various healthcare settings, such as emergency surgery patients, there is a need for rapid detection of SARS-CoV-2. The QuantuMDx Q-POC is an automated fast workflow platform that detects three genes, allowing differentiation between SARS-CoV-2 and other coronaviruses. This study compared QuantuMDx Q-POC with the Cobas 6800 algorithm for SARS-CoV-2 detection, showing high sensitivity and a short detection time of 30 minutes. Overall, the QuantuMDx Q-POC system is a reliable option for rapid SARS-CoV-2 detection.

Evaluation of Plasma Lipocalin-2 as a Predictor of Etiology and Severity in Adult Patients with Community-Acquired Pneumonia

Summary: The aim of this study was to evaluate the diagnostic performance of plasma Lipocalin-2 (LCN2) concentration in adult patients with community-acquired pneumonia (CAP) and its association with etiology, severity, and prognosis. The study found that LCN2 concentration was higher in bacterial CAP compared to non-bacterial CAP and had a limited ability to distinguish between the two. It also found that LCN2 concentration was associated with disease severity in adult patients with CAP. However, its utility as a biomarker to discriminate between viral and bacterial etiology in CAP is limited.

Summary: In recent decades, there has been an increasing trend in infections caused by multidrug-resistant (MDR) bacteria, which are associated with higher morbidity and mortality. Various interventions should be implemented to reduce the emergence and spread of MDR bacteria. This mini-review aims to compile and discuss different strategies for eradicating MDR bacteria from the gut microbiota.

Occurrence of plasmid-mediated fosfomycin resistance (fos genes) among Escherichia coli isolates, Portugal

Summary: This study evaluated the occurrence of plasmid-mediated fos genes among fosfomycin-resistant Escherichia coli isolates collected from patients in Lisbon, Portugal. The results showed a low prevalence of fosfomycin-resistant E. coli isolates, but 15% of these isolates carried transferable fosA genes, and a high rate of extended-spectrum beta-lactamase (ESBL) production was observed.

Native Pig Neutrophil Products: Insights into Their Antimicrobial Activity

Summary: This study aimed to explore the antimicrobial activity and mechanisms of action of a porcine neutrophil cathelicidin mixture (MPPN). It was found that MPPN exhibited bactericidal activity against E. coli at high concentrations and bacteriostatic activity at lower concentrations. The interaction of MPPN with lipid bilayers caused alterations in the structure.

In Vitro Effect of Three-Antibiotic Combinations plus Potential Antibiofilm Agents against Biofilm-Producing Mycobacterium avium and Mycobacterium intracellulare Clinical Isolates

Summary: Patients with chronic pulmonary diseases infected by Mycobacterium avium complex (MAC) often develop complications and suffer from treatment failure due to biofilm formation. There is a lack of correlation between in vitro susceptibility tests and the treatment of clinical isolates producing biofilm. This study found that the use of potential antibiofilm agents (PAAs) in combination with three-drug combinations can effectively reduce the bactericidal concentration in biofilm forms of MAC isolates.
