Since the discovery of unusual anti-tumor activity of natural polyether antibiotic - Salinomycin, this compound, along with its derivatives, has been intensively studied against different human cancer cells, both in vivo and in vitro. Salinomycin has shown strong inhibition activity against the proliferation process of many different cancer cells, including multi-drug resistance (MDR) cancer cells, as well as cancer stem cells (CSCs), i.e. leukemic stem cells, colon carcinoma stem cells, prostate cancer stem cells and many others. Additionally, the application of Salinomycin has been proved to enhance the anti-cancer effect of radio- and chemotherapy. Preliminary clinical studies have shown tumor regression and only transient acute side effects after application of Salinomycin. Up to now, major efforts have been devoted to elucidate the biological mechanisms of anti-tumor activity of Salinomycin and it is expected that the results may provide new therapeutic strategies based on biological modulation of Salinomycin activity. This review is focused on and describes the possible role of Salinomycin in cancer therapy and gives an overview of its properties.

Role of Vitamin E in Selected Malignant Neoplasms in Women

Anna Markowska, Michal Antoszczak, Janina Markowska, Adam Huczynski

Summary: Vitamin E, a powerful antioxidant, protects fatty acids from oxidation and has potential anticancer effects by reducing cancer stem cell activity. It is also involved in protection against mutagenic effects, anti-angiogenic activity, and inhibition of invasion and metastasis of neoplastic cells.

Synthesis and evaluation of antibacterial and trypanocidal activity of derivatives of monensin A

Marta Jedrzejczyk, Natalia Stepczynska, Greta Klejborowska, Malgorzata Podsiad, Joanna Stefanska, Dietmar Steverding, Adam Huczynski

Summary: This study presents the synthesis and biological evaluation of eleven derivatives of the natural polyether ionophore monensin A, modified at the C-26 position. These derivatives showed varying degrees of antimicrobial and antiparasitic activity. One phenyl urethane derivative exhibited the most promising antibacterial activity.

Role of Vitamin C in Selected Malignant Neoplasms in Women

Anna Markowska, Michal Antoszczak, Janina Markowska, Adam Huczynski

Summary: The effectiveness of vitamin C in fighting cancer has been widely discussed and researched. High concentrations of vitamin C have been shown to selectively destroy cancer cells, and intravenous administration of high doses of vitamin C may improve patients' quality of life. The use of vitamin C may contribute to reducing chemotherapy-related side effects.

Activity of singly and doubly modified derivatives of C20-episalinomycin against Staphylococcus strains

Dominika Czerwonka, Malgorzata Podsiad, Joanna Stefanska, Michal Antoszczak, Adam Huczynski

Summary: Chemical modification of natural polyether ionophore salinomycin has been found to be a promising strategy to generate semisynthetic analogs with improved biological activity profiles. Particularly, regioselective O-acylation of the C20-epi-hydroxyl has resulted in promising antibacterial active agents with excellent inhibitory activity against planktonic staphylococci and potential in preventing the formation of bacterial biofilms. These non-genotoxic salinomycin derivatives represent attractive candidates for further antimicrobial drug development.

Iron-Sensitive Prodrugs That Trigger Active Ferroptosis in Drug- Tolerant Pancreatic Cancer Cells

Michal Antoszczak, Sebastian Muller, Tatiana Caneque, Ludovic Colombeau, Nelson Dusetti, Patricia Santofimia-Castano, Christine Gaillet, Alain Puisieux, Juan Lucio Iovanna, Raphael Rodriguez

Summary: Persisting cancer cells are resistant to therapy and can adopt a drug-tolerant state through epithelial-mesenchymal plasticity. This study synthesized complex small molecule chimeras that accumulate in lysosomes and induce ferroptosis in drug-tolerant pancreatic cancer cells. The findings pave the way for the development of new cancer medicines targeting active ferroptosis.

Role of Vitamin K in Selected Malignant Neoplasms in Women

Anna Markowska, Michal Antoszczak, Janina Markowska, Adam Huczynski

Summary: The main function of vitamin K in the human body includes its involvement in the blood clotting cascade and its anticancer effects. Epidemiological studies have shown that reduced intake of vitamin K may increase the risk of geriatric diseases and cancer. Recent research has also indicated that vitamin K has potential in fighting against female malignancies.

Role of Hyaluronic Acid in Selected Malignant Neoplasms in Women

Anna Markowska, Michal Antoszczak, Janina Markowska, Adam Huczynski

Summary: Hyaluronic acid (HA), a significant component of the extracellular matrix, plays a crucial role in cell localization and proliferation. However, high levels of HA are associated with negative prognosis in cancer patients due to the correlation with multidrug resistance, metastasis, and cancer progression. This review summarizes the relationship between HA and the CD44 receptor, as well as other components of the HA signaling pathway, and explores their potential as prognostic biomarkers and targets for targeted therapy in gynecological cancers.

Synthesis of urea and thiourea derivatives of C20-epi-aminosalinomycin and their activity against Trypanosoma brucei

Michas Antoszczak, Kieran Gadsby-Davis, Dietmar Steverding, Adam Huczy

Summary: Salinomycin (SAL) is a natural compound with a broad spectrum of biological effects. In this study, novel urea and thiourea analogs of C20-epi-aminosalinomycin were synthesized and evaluated for their trypanocidal and cytotoxic activities. Among the tested compounds, 4b and 4d showed the most potent antitrypanosomal activity and were capable of inducing faster cell swelling in bloodstream-form trypanosomes compared to the reference compound SAL. These findings suggest that C20-epi-aminosalinomycin derivatives have the potential to be developed into improved trypanocidal drugs.

The Role of Selected Dietary Factors in the Development and Course of Endometriosis

Anna Markowska, Michal Antoszczak, Janina Markowska, Adam Huczynski

Summary: Endometriosis is a chronic disease that affects about 10% of women of reproductive age, causing pain and infertility. Treatment includes medication and surgical removal of lesions, but recurrence rates remain high. This review article focuses on the potential benefits of polyphenols, vitamins, and micronutrients in fighting endometriosis. Although studies have shown promising results in animal models, more well-designed studies are needed to evaluate the effects of dietary factors on women with endometriosis.
