Summary: New liposomes modified with pyrrolidinium surfactants containing a hydroxyethyl fragment were prepared for transdermal delivery of non-steroidal anti-inflammatory drugs. Liposome modification led to prolonged release of rhodamine B and faster release of non-steroidal drugs in vitro, with C16PB-modified liposomes found to be the most effective in reducing inflammation in a rat paw edema model.

Summary: In response to the global need for an effective vaccine in the SARS-CoV-2 pandemic, a plasmid DNA vaccine expressing the spike (S) protein was developed and tested for in vitro transfection and in vivo immunogenicity. The formulated liposomes complexed with the DNA vaccine showed enhanced neutralizing antibody responses and T cell responses, comparable to the efficacy of intramuscular electroporation (IM-EP) as a benchmark.

Summary: We improved the bioavailability of carvedilol (CVD) by fabricating intranasal administration of CVD-loaded cationic nanoliposomal (NLPs) in situ gel. The CVD NLPs in situ gel showed a 6.94-fold increase in Cmax and a 5-fold increase in relative bioavailability compared to an oral drug suspension, indicating the potential of cationic NLPs as an alternative carrier for intranasal administration.

Summary: This study presents an extension of the Membrane Builder tool to model and simulate lipid nanoparticles (LNPs) with various lipid types and PEG-lipids effectively. The capability of Membrane Builder to construct realistic LNP membranes with high concentrations of cholesterol, ionizable cationic lipids, and PEG-lipids is demonstrated, showing the flexibility of PEG-chains and the effects of PEG-lipids on lateral packing and membrane properties. The interactions between PEG oxygen and the head group of ionizable cationic lipids induce negative curvature, which could be useful for better characterizing different LNPs for rational LNP design.
