Summary: Our understanding of the identity of many cancers has greatly increased in recent years, leading to progress in early detection and treatment options. However, gastric cancer remains poorly treated with low survival rates. The lack of possibilities for early detection and the variations between tumors of gastric cancer patients are major challenges. Histone Deacetylases (HDACs) have been identified to be potentially related to gastric cancer. In this review, we summarize the current knowledge on the role of HDACs in gastric cancer development and their potential as early detection markers and targets for new treatment options.

Summary: This study demonstrates that the expression of lncH19 contributes to ITF2357-induced apoptosis by stabilizing TP53. Overall, we suggest that lncH19 expression may be exploited to enhance HDACi-induced cell death and overcome 5-fluorouracil chemoresistance.

Summary: Cancer is the second leading cause of death worldwide, with digestive system cancers being a primary contributor. Acetylation and deacetylation play crucial roles in cancer development, with HDAC6 being a widely studied enzyme that is upregulated in various tumors and associated with clinicopathological characteristics. There is ongoing research on HDAC6 inhibitors and their potential in inhibiting tumor growth.

Summary: Cerebral ischemia is the second leading cause of death worldwide, requiring multimodal stroke therapy. Histone deacetylase inhibitors have shown to be effective in protecting the brain from ischemic damage by inducing neurogenesis and angiogenesis in damaged brain areas, promoting functional recovery after stroke.

Summary: Histone deacetylases (HDACs) can deacetylate histones H3 and H4, and an imbalance in histone acetylation and deacetylation can contribute to various diseases. HDAC2, located in the nucleus, plays a critical role in modifying chromatin structures and regulating gene expression as a transcriptional regulator. This review discusses the roles of HDAC2 in tumorigenesis and anti-cancer drug resistance, and highlights its potential as a prognostic marker for various cancers. It also explores the involvement of microRNAs in directly regulating the expression of HDAC2 during tumorigenesis, and emphasizes HDAC2 as a valuable target for developing anti-cancer drugs.

Summary: Histone deacetylase inhibitors (HDACIs) have important effects on epigenetic regulation in cancer, including inducing cancer cell death, apoptosis, and cell cycle arrest. Although mainly approved for hematologic malignancies, there are promising studies in cervical cancer. This comprehensive review examines the role of HDACIs in the treatment of cervical cancer and discusses their potential as effective anticancer drugs, either alone or in combination with other treatments.

Summary: Immunotherapy with checkpoint inhibitors has replaced chemotherapy in some settings, allowing many patients to live longer with fewer side effects, but only works for about one-fifth of unselected patients. Important cofactors within the tumor microenvironment, such as T-cells and chemokines, can impact the efficacy of immunotherapy through epigenetic modulation.

Summary: Post-translational histone modifications, such as histone acetylation, play key roles in regulating gene expression in plant growth, development, and stress responses. The research found that HDAC inhibitors led to hyperacetylation of histone H3, enhancing MeJA-induced ginsenoside production in ginseng adventitious roots. Additionally, the study identified specific PgHDACs that may serve as crucial factors in controlling MeJA-induced ginsenoside production.

Summary: This study investigated the influence of histone acetylation on secondary metabolism in Platycodon grandiflorus by treating it with sodium butyrate (NaB), a histone deacetylase inhibitor. The treatment increased histone acetylation and enhanced the anti-melanogenic properties of P. grandiflorus roots. The accumulation of polyphenolic compounds and the activation of genes in the phenylpropanoid biosynthetic pathway were found to be responsible for the enhanced medicinal qualities.

Summary: Myeloid-derived suppressor cells (MDSCs) are heterogeneous populations that suppress tumor-specific T-cell responses and are increased in several hematologic malignancies, including chronic myeloid leukemia (CML). In CML patients, differences in levels and prognostic significance of MDSCs are evaluated according to clinical status. High levels of peripheral blood granulocytic MDSCs (gMDSCs) are associated with better prognosis, while monocytic MDSCs (mMDSCs) may be related to malignant conditions and poor prognosis.

Summary: Primary graft failure (GF) has a high mortality rate, while the 10-year cumulative incidence of secondary GF is 4.5%, with a median onset of 3 months post-transplant and a 5-year Kaplan-Meier survival of 28.8%. The diagnosis of severe aplastic anemia and use of reduced intensity conditioning were both associated with higher incidence and better survival rates in the secondary GF group.
