Bacillus ligniniphilus sp nov., an alkaliphilic and halotolerant bacterium isolated from sediments of the South China Sea

An alkaliphilic and halotolerant Gram-stain-positive bacterium, which was isolated from sediment samples from the South China Sea, was subjected to a taxonomic study. The isolate, strain L1(T), grew well at a wide range of temperatures and pH values, 10.0-45.0 degrees C and pH 6-11, with optima at 30 degrees C and pH 9.0, respectively. The growth of strain LIT occurred at total salt concentrations of 0-10% (w/v) with an optimum at 2% (w/v). Phylogenetic analysis based on 16S rRNA sequence comparison indicated that the isolate represented a member of the genus Bacillus. The strains most closely related to strain L1(T) were Bacillus nanhaiisediminis JCM 16507(T), Bacillus halodurans DSM 497(T) and Bacillus pseudofirmus DSM 8715(T), with 16S rRNA similarities of 96.5 %, 95.9% and 95.7%, respectively. DNA DNA hybridization of strain L1(T) with the type strains of the most closely related species, B. nanhaiisediminis JCM 16507(T), B. halodurans DSM 497(T) and B. pseudofirmus DSM 8715(T), showed reassociation values of about 21.7%, 14.3% and 13.9 %, respectively. The DNA G+C content of strain L1(T) was 40.76 mol%. The predominant isoprenoid quinone was menaquinone 7 (MK-7). The cell-wall peptidoglycan contained meso-diaminopimelic acid as the diagnostic diamino acid. The predominant cellular fatty acids of strain L1(T) were iso-C-14:0, and anteiso-C-15:0. The major polar lipids were diphosphatidylglycerol, phosphatidylethanolamine and phosphatidylglycerol. Based on the phenotypic and phylogenetic characteristics, it is proposed that strain L1(T) (=JCM 18543(T)=DSM 26145(T)) should be classified as the type strain of Bacillus ligniniphilus sp. nov.

Microbial diversity and community structure in deep-sea sediments of South Indian Ocean

Summary: In this study, microbial diversity, community composition and dynamics in microbial community structure in deep-sea sediments of the South Indian Ocean were explored using Next-Generation Sequencing. The results showed that Proteobacteria and Firmicutes were the dominant bacterial phyla, while Thaumarchaeota and Euryarchaeota were the abundant archaeal phyla. Ascomycota and Basidiomycota were the most abundant fungal phyla. It was also found that bacterial communities tended to be more dynamic than archaeal and fungal communities.

Self-Assembled Microfiber-Like Biohydrogel for Ultrasensitive Whole-Cell Electrochemical Biosensing in Microdroplets

Summary: A novel microfiber-like biohydrogel was developed using electroactive bacteria-induced graphene oxide reduction and confined self-assembly in a capillary tube. This biohydrogel enabled efficient electron exchange between cells and the conductive graphene network, and was used to create a miniature whole-cell electrochemical biosensing system for fumarate detection. The system exhibited high sensitivity and a low limit of detection, offering new possibilities for bioelectrochemical detection in small volumes or at high spatial resolutions.

Straw lignin degradation by lignin peroxidase from Irpex lacteus cooperated with enzymes and small molecules

Summary: By studying the effects of small molecules on the efficiency of lignin degradation by lignin peroxidase, the utility value of enzymes was maximized. Using wheat straw as raw material, it was found that laccase, glucose oxidase, malonic acid, citric acid, ZnSO4, CaCl2 could promote the lignin degradation by the lignin peroxidase. Moreover, glucose oxidase, malonic acid and CaCl2 had obvious synergy effects on lignin degradation by the lignin peroxidase.

Biodegradability of polyethylene by efficient bacteria from the guts of plastic-eating waxworms and investigation of its degradation mechanism

Summary: This study investigated the potential of gut symbionts of lesser waxworm larvae to degrade low-density polyethylene (LDPE). Two bacterial isolates, identified as Citrobacter freundii and Bacillus sp., showed the highest reduction in LDPE's tensile strength. The cell density of these bacteria increased over time, indicating their high activity in degrading LDPE. These findings provide promising evidence for the biodegradation of plastic waste management in the environment.

A review of microbial degradation of per- and polyfluoroalkyl substances (PFAS): Biotransformation routes and enzymes

Summary: Since the 1950s, large amounts of PFAS have been dumped into the environment, causing significant contamination. PFAS are resistant to degradation and have adverse effects on health and the environment. Traditional physicochemical methods for PFAS treatment have drawbacks, making microbial degradation and enzymatic treatment highly sought after. This review provides an overview of PFAS classification, properties, and interaction with the environment, and discusses recent developments in microbial degradation, transformation routes, and challenges in bioremediation approaches.

Characterization of the Genetic Variability within Ziziphus nummularia Genotypes by Phenotypic Traits and SSR Markers with Special Reference to Geographic Distribution

Summary: Understanding the impacts and constraints of climate change on the geographical distribution of Ziziphus nummularia is crucial for its sustainability. This study analyzed field investigation and climate change data using ArcGIS analysis. Genetic diversity of 180 genotypes from three populations was studied using morphological attributes and simple sequence repeat (SSR). Results showed that mean temperature was a significant factor limiting the distribution. Current and future suitable regions were identified as Swat and Buner, respectively. Phenotypic traits and genetic diversity were assessed, indicating the presence of variability and unique alleles. Conservation strategies are required due to the impact of climate change on species distributions.

Astaxanthin production from the microalga Haematococcus lacustris with a dual substrate mixotrophy strategy

Summary: This study investigates the development of a dual-substrate mixotrophy strategy to cultivate the microalga Haematococcus lacustris for astaxanthin production. The results showed that the dual-substrate mixotrophy significantly increased biomass productivity by up to 2-fold in the green growth phase compared to phototrophic controls. Furthermore, the supplementation of dual-substrate to the red phase increased astaxanthin accumulation by 10% compared to single-substrate acetate and no substrate. This approach shows promise for commercial production of biological astaxanthin in indoor closed systems.

The role of microorganisms in petroleum degradation: Current development and prospects

Summary: Petroleum hydrocarbon compounds are persistent organic pollutants that can cause permanent damage to ecosystems due to their biomagnification. Bioremediation is currently the main solution for remediation of petroleum hydrocarbon pollutants in ecosystems. However, there are various challenges for microorganisms to perform efficiently in outside environments, making it necessary to explore efficient biodegradation technologies through the discovery of new microorganisms, modification of biodegradation pathways, and development of new bioremediation technologies.

Immediate health risk: Concentration of heavy metals in contaminated freshwater fishes from the river channel of Turag-Tongi-Balu

Summary: The consumption of contaminated finfish from the polluted river of Turag-Tongi-Balu, Kamarpara site, Dhaka poses significant health hazards to humans, with the concentrations of Cd, Mn, Pb, and Se exceeding the Food Safety Guideline value in most fish species. The high Mn concentrations in certain fish species elevate the non-carcinogenic risk, and there are statistically significant differences in heavy metal concentrations among fish species. The regular dietary consumption of fish from this polluted ecosystem poses a significant health risk and is indicated as cancer, emphasizing the importance of monitoring heavy metal contamination and minimizing the risk to human health.

Green alkaline fractionation of sugarcane bagasse at cold temperature improves digestibility and delignification without the washing processes and release of hazardous waste

Summary: A response surface design was used to pretreat sugarcane bagasse (SB) with urea (U) and potassium hydroxide (K) at low temperatures. The optimized pretreatment condition (-20 degrees C, 7.5% UK) resulted in increased removal of hemicellulose and lignin, as well as an increase in cellulose content compared to the control. The UK pretreatment also improved enzymatic hydrolysis and exhibited potential as a pretreatment technology for lignocellulosic valorization.

Optimization of heterologous production of Bacillus ligniniphilus L1 laccase in Escherichia coli through statistical design of experiments

Summary: This study optimized the production of bacterial laccase, a promising enzyme for lignin valorization and biomass processing, by improving culture medium composition and fermentation parameters. The optimized medium achieved a 3.3-fold yield improvement, and further optimization of fermentation parameters resulted in a 7-fold yield increase compared to initial conditions. This research provides a high-yielding and cost-efficient production system for bacterial laccase and has implications in various applications.

Lignin-based additive materials: A review of current status, challenges, and future perspectives

Summary: Additive manufacturing, particularly using renewable natural polymers and synthetic materials, shows great potential in the manufacturing, pharmaceutical, food, and biomedical industries. Among these materials, lignin, which is a renewable bio-based raw material, is receiving significant attention due to its amorphous macromolecular structure and beneficial properties. However, the complex polymeric and amorphous structure of lignin presents challenges for its utilization in 3D printing. This review summarizes the use of lignin in the preparation of ink for 3D printing and discusses its merits, limitations, and future perspectives.

Bioremediation of Azo Dye Brown 703 by Pseudomonas aeruginosa: An Effective Treatment Technique for Dye-Polluted Wastewater

Summary: Dye-polluted wastewater is a serious threat to human, animal, and plant health. To mitigate this, the treatment of wastewater containing dyes is necessary. This study demonstrates the use of Pseudomonas aeruginosa to degrade the textile azo dye brown 703. Optimization experiments showed that under specific conditions, the bacterial strain can effectively decolorize and degrade the dye within 3 days.
