Gram-positive, endospore-forming, rod-shaped bacterium, designated isolate J4(T), was isolated from a neutral saline lake sample from Xinjiang Uyghur Autonomous Region, China, and subjected to a polyphasic taxonomic investigation. Phylogenetic analysis based on 16S rRNA gene sequence revealed that strain J4(T) is most closely related to Salinibacillus aidingensis 25-7(T) (with 96.7% similarity), Salinibacillus kushneri 8-2(T) (96.5%), Ornithinibacillus scapharcae TW25(T) (96.4 %), Salirhabdus euzebyi CVS-14(T) (96.4 %) and Ornithinibacillus californiensis MB-9(T) (96.2%). Chemotaxonomic analysis showed menaquinone-7 (MK-7) to be the major isoprenoid quinone of strain J4(T); diphosphatidylglycerol and phosphatidylglycerol were the major cellular polar lipids and the cell wall contained meso-diaminopimelic acid as the diagnostic diamino acid. The major cellular fatty acids were iso-C-(15:0) and anteiso-C-15:0. The genomic DNA G+C content of strain J4(T) was determined to be 36.2 mol%. Strain J4(T) was positive for catalase activity and negative for oxidase activity. Strain J4(T) was observed to grow at 25-50 degrees C (optimal 35-42 degrees C), pH 6.5-8.0 (optimal 7.0-7.5) and in media containing 1-21% (w/v) NaCl (optimal 9-12%). Based on these data, strain J4(T) represents a novel species of the genus Salinibacillus and the name Salinibacillus xinjiangensis sp. nov. is proposed. The type strain is J4(T) (=CGMCC 1.12331(T)=JCM 18732(T)).

Natural products from traditional medicine as promising agents targeting at different stages of oral biofilm development

Yaqi Chi, Ye Wang, Mengzhen Ji, Yanyao Li, Hualing Zhu, Yujia Yan, Di Fu, Ling Zou, Biao Ren

Summary: The oral cavity is a habitat for over 1,000 species of microorganisms, which form biofilms and impact oral ecological balance and the development of oral diseases. Natural products from traditional medicine have shown potential antibiofilm activities and can be safe and cost-effective. Therefore, it is important to study and develop these natural products as antibiofilm agents for the oral cavity.

Candida albicans CHK1 gene regulates its cross-kingdom interactions with Streptococcus mutans to promote caries

Yaqi Liu, Zheng Wang, Ziyi Zhou, Qizhao Ma, Jing Li, Jun Huang, Lei Lei, Xuedong Zhou, Lei Cheng, Jing Zou, Biao Ren

Summary: This study demonstrated that Candida albicans CHK1 gene plays a crucial role in the development of dental caries by promoting the growth, biofilm formation, and exopolysaccharides production of Streptococcus mutans. The upregulation of S. mutans vicR and vicK genes by C. albicans CHK1 gene enhances the expression of EPS biosynthesis genes, leading to increased EPS production and biofilm formation. In a rat caries model, coinfection with chk1 Delta/Delta and S. mutans resulted in decreased colonization of S. mutans and less severe caries compared to other combinations.

The Preventive Effect of A Magnetic Nanoparticle-Modified Root Canal Sealer on Persistent Apical Periodontitis

Xiao Guo, Yan Sun, Zheng Wang, Biao Ren, Hockin H. K. Xu, Xian Peng, Mingyun Li, Suping Wang, Haohao Wang, Yao Wu, Michael D. Weir, Xuedong Zhou, Fang Lan, Lei Cheng

Summary: Persistent apical periodontitis is a critical challenge in root canal therapy. Developing root canal filling materials with continuous antibacterial effects and tight seal is important to prevent treatment failure and persistent apical periodontitis. In this study, we modified the EndoREZ root canal sealer by adding the antibacterial material DMADDM and magnetic nanoparticles (MNPs). The modified sealer showed good mechanical properties, biocompatibility, and enhanced antibacterial effects on multispecies biofilms under a magnetic field. In vivo study demonstrated its inhibitory effect on persistent apical periodontitis with minimal resorption and slight increase in periodontal ligament space.

The Bio-Aging of Biofilms on Behalf of Various Oral Status on Different Titanium Implant Materials

Min Liao, Yangyang Shi, Enni Chen, Yuke Shou, Dongyue Dai, Wenpan Xian, Biao Ren, Shimeng Xiao, Lei Cheng

Summary: This study investigated the effects of bio-aging on titanium implants in the oral microenvironment. Different surface treatments were used, and biofilms were established using different bacteria and saliva from healthy individuals and patients with periodontitis. After 30 days of bio-aging, the surface morphology, chemical composition, and water contact angles were measured. The adhesion of cells and biofilms was also evaluated. The results showed that the polished samples had no significant changes, while the MAO- and SLA-treated samples showed mild corrosion. The HA-coated samples were the most vulnerable, especially when exposed to saliva from patients with periodontitis. These findings highlight the importance of oral hygiene and periodontal health for long-term implant success.

Correspondence on Structural Insight into the Catalytic Mechanism of the Endoperoxide Synthase FtmOx1

Lixin Zhang, Xueting Liu, Xinye Wang, Guoliang Zhu, Heng Song, Ronghai Cheng, Nathchar Naowarojna, Catherine E. E. Costello, Pinghua Liu

Summary: In their recent publication, Cen, Wang, Zhou et al. reported the crystal structure of a ternary complex of the non-heme iron endoperoxidase FtmOx1 and concluded that it supports the CarC-like mechanistic model for catalysis. However, they failed to accurately describe the CarC-like and COX-like models and address their differences, and their data is consistent with the COX-like model after careful analysis.

OvoAMtht from Methyloversatilis thermotolerans ovothiol biosynthesis is a bifunction enzyme: thiol oxygenase and sulfoxide synthase activities

Ronghai Cheng, Andrew C. Weitz, Jared Paris, Yijie Tang, Jingyu Zhang, Heng Song, Nathchar Naowarojna, Kelin Li, Lu Qiao, Juan Lopez, Mark W. Grinstaff, Lixin Zhang, Yisong Guo, Sean Elliott, Pinghua Liu

Summary: This work reports the dual activities of a non-heme iron enzyme OvoA(Mtht) in Methyloversatilis thermotolerans, acting as a thiol oxygenase and a sulfoxide synthase. The control of the enzyme's dual activities by the substrates and the active site iron's secondary coordination shell residues is demonstrated. This finding provides an excellent system for future mechanistic investigation on the modulation of iron-centers' electronic properties for different reactivities.
