Allonocardiopsis opalescens gen. nov., sp nov., a new member of the suborder Streptosporangineae, from the surface-sterilized fruit of a medicinal plant

An endophytic actinomycete, designated strain I1A-0-1259(T), was isolated from a surface-sterilized fruit of Lonicera maackii (Rupr.) Maxim., a medicinal plant, which was collected from a suburb of Beijing, China. Whole-cell hydrolysates of the isolate contained galactose and meso-diaminopimelic acid. The predominant phospholipids were phosphatidylglycerol and diphosphatidylglycerol; the menaquinones consisted mainly of MK-9, MK-11 and MK-12, with a minor amount of MK-10. The major fatty acids were iso-C-16:0, anteiso-C-15:0 and iso-C-15:0. Comparative analysis of 16S rRNA gene sequences indicated that strain I10A-01259(T) was most closely related to Nocardiopsis arable S186(T) (93.2% sequence similarity), Thermobifida halotolerans YIM 90462(T) (93.0%) and other strains of genera within the families Nocardiopsaceae and Thermomonosporaceae. On the phylogenetic tree based on 16S rRNA gene sequences, strain I10A-01259(T) fell within the radius of the suborder Streptosporangineae, in which the strain formed a distinct lineage next to the genera of the families Nocardiopsaceae and Thermomonosporaceae. Based on the data from our polyphasic taxonomic study, a novel genus and species, Allonocardiopsis opalescens gen. nov., sp. nov., are proposed within the suborder Streptosporangineae. The type strain of Allonocardiopsis opalescens is strain I10A-01259(T) (=CPCC 203428(T) = DSM 45601(T) = KCTC 19844(T)).

Summary: This study aims to discover new drugs targeting the virulence factor ESX-1 in Mycobacterium tuberculosis. Using a high-throughput screening assay, a compound named IMB-BZ was identified as a specific inhibitor of ESX-1 secretion, showing significant antibacterial and virulence-reducing effects. Importantly, IMB-BZ exhibited low risk of inducing drug resistance in vitro and has the potential to be developed into a novel anti-TB drug.

Metagenomic analysis reveals microbiome and resistome in the seawater and sediments of Kongsfjorden (Svalbard, High Arctic)

Summary: This study analyzed the diversity and distribution of bacterial communities and antibiotic resistance genes (ARGs) in seawater and sediments of Kongsfjorden in the Arctic through metagenomic analysis. The results showed significant differences in bacterial community compositions and ARG profiles between seawater and sediment samples. Geographic and geochemical factors greatly influenced the bacterial communities and ARG profiles, with Procrustes analysis revealing a significant correlation between the two.

Discovery and Activation of the Cryptic Cluster from Aspergillus sp. CPCC 400735 for Asperphenalenone Biosynthesis

Summary: This study successfully manipulated the fungus Aspergillus sp. CPCC 400735 to activate the downregulated metabolic pathway encoded by the biosynthetic gene cluster asp through overexpression of the transcriptional regulator AspE. 13 asperphenalenone derivatives, including 11 new compounds, were isolated and characterized from the mutant extracts, showing significant anti-influenza A virus effects with IC values of 0.45-2.22 μM. The identification of these compounds sheds light on the biosynthesis of asperphenalenones and could benefit downstream combinatorial biosynthesis studies.

Summary: Researchers have discovered a compound called APL-16-5 that exhibits antiviral effects against influenza A virus (IAV). APL-16-5 binds to TRIM25 and viral PA protein, leading to the degradation of PA through ubiquitination and proteasome-mediated pathways. This compound shows potent in vivo anti-IAV activity and has the potential to become a new therapeutic option for IAV.

Shinella lacus sp. nov., a novel microcystin- degrading alphaproteobacterium containing the bla carbapenemase gene

Summary: A microcystin-degrading bacterium, Shinella lacus sp. nov., was isolated from a fresh water reservoir in Sichuan Province, China. It exhibited optimal growth at 28-32 degrees C and pH 7.0, and contained genes encoding peroxidase, bla carbapenemase, and the key enzyme for microcystin bio degradation. The bacterium showed adaptability to diverse environments.

A New Isobenzofuranone Derivative from Arctic Fungus Gyoerffyella sp. CPCC 401434

Summary: A new isobenzofuranone derivative and six known compounds were isolated from Arctic fungus Gyoerffyella sp. CPCC 401434, and their structures and absolute configurations were determined. This is the first report of these compounds from the genus Gyoerffyella, and their cytotoxic and antibacterial activities were also studied.

An Integrative Study of Mycobiome in Different Habitats from a High Arctic Region: Diversity, Distribution, and Functional Role

Summary: In Arctic ecosystems, fungi play a crucial role in soil-plant interactions, nutrient cycling, and carbon transport. However, the mycobiome and its functional role in different habitats of the High Arctic region have not been thoroughly studied. This research aimed to investigate the mycobiome in nine habitats using high-throughput sequencing and found a total of 10,419 detected ASVs. The distribution of the mycobiome was influenced by habitat specificity, and various ecological guilds and growth forms were identified among different habitats.

Summary: This study reveals the genome and the ability of Desarmillaria tabescens CPCC 401429 fungus to produce diverse sesquiterpene compounds. Through genomic analysis and transcriptomic sequencing, researchers have identified some novel sesquiterpene synthase genes and characterized two of them, confirming the potential of Desarmillaria in generating novel terpenoids.

Shotgun metagenomics reveals a diverse mycobiome in the seawater from a High Arctic fjord (Kongsfjorden, Svalbard)

Summary: This study comprehensively characterized the mycobiome in seawater samples from Kongsfjorden, Svalbard, using shotgun metagenomics. The results showed a diverse mycobiome with significant differences in composition among different water layers. Depth, NO2 , and PO43  were identified as key factors influencing the mycobiome composition.

Verbalide A∼F: new phthalide derivatives from the endophytic fungus Preussia sp. CPCC 400972

Summary: Six new phthalide derivatives, namely Verbalide A similar to F (1-6), and one known derivative (7), were isolated from the endophytic fungus Preussia sp. CPCC 400972. The structures of these compounds were determined through comprehensive spectroscopic analyses, including NMR and HRESIMS. Furthermore, compounds 1-7 showed excellent inhibitory effect against influenza A virus.

Phenol Derivatives With Antiviral Activities from the Arctic Fungus Phoma muscivora CPCC 401424

Summary: A new phenol derivative (1) and seven known phenol analogs (2-8) were isolated from Arctic fungus Phoma muscivora CPCC 401424. Their structures were determined using spectroscopic techniques and comparing with existing data. Compound 1 and 5 exhibited strong anti-influenza A virus activities with IC50 values of 2.20 and 2.17 μM, respectively. Compounds 1, 2, 5, and 7 showed moderate cytotoxic activities against Hela and H460 cancer cell lines.

New dimeric chromanone derivatives from the mutant strains of Penicillium oxalicum and their bioactivities

Summary: Three new chromanone dimer derivatives, paecilins F-H (1-3), were isolated from mutant strains of Penicillium oxalicum 114-2. Their structures were elucidated and their configurations were resolved. Compounds 5 and 11 showed significant anti-influenza A virus activities, compounds 8 and 9 exhibited cytotoxic activities against the MIA-PaCa-2 cell line, and compound 10 showed antibacterial activities against Bacillus cereus.
