Jatrophihabitans endophyticus gen. nov., sp nov., an endophytic actinobacterium isolated from a surface-sterilized stem of Jatropha curcas L.

A short rod-shaped Gram-stain-positive actinobacterium was isolated as an endophyte from the tissues of Jatropha curcas cv. KB27 and was investigated by means of a polyphasic taxonomic approach. An analysis of its 16S rRNA gene sequence indicated that strain S9-650(T) forms an individual line of descent and is related to certain members of the suborder Frankineae, order Actinomycetales (<95 % sequence similarity). Distance-matrix and neighbour-joining analyses set the branching point of the novel isolate between two clades, one being represented by members of the genera Frankia (family Frankiaceae) and Acidothermus (family Acidothermaceae) and the other by members of the genera Geodermatophilus, Blastococcus and Modestobacter (family Geodermatophilaceae). The organism had meso-diaminopimelic acid as the diagnostic diamino acid in the cell-wall peptidoglycan. The acyl type was found to be N-glycolylated. The major menaquinone was MK-9(H-4) and the fatty acid profile was characterized by the predominance of iso-C-16:0, C-18:1 omega 9c, anteiso-C-17:0 and C-17:1 omega 8c. The polar lipids comprised diphosphatidylglycerol, an unidentified glycolipid, phospholipids and aminolipids. The G+C content of the genomic DNA was 71.2 mol%. The distinct phylogenetic position and the phenotypic markers that clearly separate the novel organism from all other members of the suborder Frankineae indicate that strain S9-650(T) represents a novel species in a new genus, for which the name Jatrophihabitans endophyticus gen. nov., sp. nov. is proposed. The type strain of the type species is S9-650(T) (=DSM 45627(T)=KACC 16232(T)).

Summary: This study investigated the diversity, antimicrobial, and cytotoxic activities of fungal endophytes obtained from different habitats in Singapore. The majority of the identified strains belonged to the phylum Ascomycota, and the most dominant genus was Colletotrichum. Chemical elicitation and variation of fermentation media led to the discovery of more bioactive strains. Active constituents were isolated and characterized, including a known compound and five novel compounds. These compounds showed strong antifungal and antiproliferative activities.

Diversity and Biosynthetic Potential of Fungi Isolated from St. John's Island, Singapore

Chemical elicitation as an avenue for discovery of bioactive compounds from fungal endophytes

Summary: This study investigated the molecular phylogeny, antimicrobial and cytotoxic activities of fungal endophytes obtained from the A*STAR Natural Organism Library (NOL) and previously isolated from Sungei Buloh Wetland Reserve, Singapore. The results showed that chemical elicitation can increase the number of active strains and new compounds with antifungal and cytotoxic activities were discovered.

ACC deaminase producing endophytic bacteria enhances cell viability of rice (Oryza sativa L.) under salt stress by regulating ethylene emission pathway

Summary: This study evaluates the efficacy of ACC deaminase producing methylotrophic bacteria in enhancing cell viability of rice roots under salt stress. The results show that Methylobacterium oryzae strain CBMB20 enhances cell viability by regulating ethylene emission pathway. Additionally, the study reveals that senescence-associated protein (SAP) mRNA expression and enzyme activity are coordinated with major phytohormones. This research provides a mechanistic understanding of the plant-microbe interactions important for stress alleviation, where Methylobacterium spp. plays a central role in enhancing plant biomass through restriction of apoptosis via regulation of ethylene emission.
