Severely Impaired and Dysregulated Cytochrome P450 Expression and Activities in Hepatocellular Carcinoma: Implications for Personalized Treatment in Patients

This study aims to systematically determine the activities and expressions of cytochrome P450s (CYP) in hepatocellular carcinoma (HCC) patients to support their optimal use in personalized treatment of HCC. Activities of seven major drug-metabolizing CYP enzymes (CYP1A2, 2A6, 2C8, 2C9, 2D6, 2E1, and 3A4) were determined in tumors and pericarcinomatous tissues harvested from 26 patients with hepatitis B virus-positive HCC using probe substrates. Protein and mRNA levels of these CYPs were also measured using isotope label-free LC/MS-MS method and real-time PCR, respectively. Maximal metabolic velocity (V-max) of CYP probe substrates was decreased by 2.5- to 30-fold in tumor microsomes, accompanied by a corresponding decrease in their protein and mRNA expression levels. However, Km values and turnover numbers of substrates in tumor microsomes were not changed. High correlations between activities and CYP protein levels were also observed, but the correlation between activities and mRNA levels was often poor. There was a major decrease in the degree of correlation in CYP expression in tumor tissues, suggesting that CYP expression levels are greatly disrupted by the tumorigenic process. Our unprecedented systemic study of the effects of HCC on CYPs demonstrated that activities of CYPs were seriously impaired and their expression patterns were severely altered by HCC. We proposed that determination of the CYP protein expression profile by LC/MS-MS in each patient is a promising approach that can be clinically used for individualized treatment of HCC. Novel treatment for refractory rheumatoid arthritis with total glucosides of paeony and nobiletin codelivered in a self-nanoemulsifying drug delivery system

Summary: This study developed a novel treatment for refractory rheumatoid arthritis by co-delivering total glucosides of paeony and the P-gp inhibitor nobiletin in a nano-SNEDDS formulation, which significantly increased the bioavailability of TGPs and inhibited P-gp function and expression. The co-loaded formulation not only enhanced the therapeutic effects of TGP but also reduced P-gp expression in an adjuvant-induced arthritis rat model, partly through the inhibition of phosphorylated AKT and HIF-1 alpha pathways.

Rationally designed novel multifunctional poly(ionic liquid)s for ultra-selective valorization of Yiwu lignite to monocyclic aromatic compounds

Summary: This study successfully synthesized hydrophobic mesoporous poly(ionic liquid)s (PILs) with strong Bronsted-Lewis acid sites and demonstrated their efficiency as catalysts for the selective valorization of lignite into monocyclic aromatic compounds (MCACs) with a yield of 23.5 wt.%. The synergistic catalysis mechanisms include the hydrophobic micro-environment created by the PILs, the mesoporous confinement that enhances substrate concentration, and the active hydrogen transfer of the Bronsted-Lewis acid sites.

A Systematic Review and Meta-Analysis of the Efficacy and Safety of Xinbao Pill in Chronic Heart Failure

Summary: This study demonstrated that Xinbao pill as an adjunctive treatment for chronic heart failure showed efficacy in improving left ventricular function, increasing total effective rate, reducing cardiac load, and improving exercise capacity with good safety profile. However, further well-designed trials are needed to confirm its therapeutic efficacy and safety due to the poor quality of included studies.

Exploring the catalytic function and active sites of a novel C-glycosyltransferase from Anemarrhena asphodeloides

Summary: Anemarrhena asphodeloides, a popular medicinal herb in China, contains mangiferin which has various pharmacological activities. A promiscuous C-glycosyltransferase (AaCGT) was identified from Anemarrhena asphodeloides, which was capable of catalyzing mono-C-glycosylation and di-C-glycosylation reactions. AaCGT showed potential as a biocatalyst for synthesizing high-value C-glycosides.
