Temoporfin-loaded liposomal gels: Viscoelastic properties and in vitro skin penetration

Temoporfin (mTHPC) is a potent second-generation photosensitizer. The primary object of this study was to develop a topical mTHPC-loaded liposomal hydrogel able to deliver mTHPC into the stratum corneum (SC) and deeper skin layers. This study was conducted (1) to determine the effect of carbomer concentration, used as a gelling agent, and the effect of phosphatidylcholine (PC) content of lecithin, used for the liposome preparation. on viscoelastic properties and viscosity of liposomal gels and (2) to determine the relationship between theological properties of gels and the skin penetration of mTHPC. Liposomal hydrogels revealed plastic flow behaviour. The increase of carbomer concentration induced a domination of elastic over viscous behaviour of gels. There was an inverse relationship between the elasticity of gels and mTHPC-penetration. Viscosity also increased with the increment of carbomer concentration, reducing the mTHPC-penetration. Liposomal gels containing lecithin of smaller PC-content (i.e. smaller purity) exhibited a more elastic solid behaviour than gels containing lecithin with high PC-content, and showed smaller mTHPC-penetration. The gel containing 0.75%, w/w, carbomer and lecithin with high PC-content was considered to be the optimal formulation, since it delivered high amounts of mTHPC to the SC and deeper skin layers, and it possessed desirable theological properties. Summary: The natural kaolin was modified with different amounts of ODMBA to create organokaolinites, and their adsorption of mycotoxins OCHRA and ZEN was studied at varying pH levels. The adsorption capacities of both mycotoxins increased with higher levels of surfactant on the kaolin surface, but varied at different pH values.

Safe-by-design gelatin-modified zinc oxide nanoparticles

Summary: An innovative low-cost wet precipitation synthesis method was reported for gelatin-modified zinc oxide nanoparticles. The nanoparticles showed slight reduction in cell viability and low genotoxic potential at very high doses in human hepatocellular carcinoma cell line HepG2. Further studies are warranted for their potential applications in antibacterial agents or dietary supplements with low overall toxicity.

Formulation of olopatadine hydrochloride viscous eye drops - physicochemical, biopharmaceutical and efficacy assessment using in vitro and in vivo approaches

Summary: This work formulated viscous eye drops using medium chain chitosan and hydroxypropyl guar gum as carriers for olopatadine. Physicochemical properties of the formulations were satisfactory, with in vitro studies showing non-toxicity and good anti-pruritic effects on eye cells. The viscous ophthalmic vehicle may be a feasible strategy to enhance the efficacy of olopatadine.

Summary: This study aimed to investigate the loading of sildenafil citrate in three commercial transdermal vehicles using 3D full-thickness skin equivalent and compare the results with permeability experiments using porcine skin. The results showed that the results obtained using the 3D skin equivalent were comparable to those obtained using porcine skin, suggesting that the 3D skin model can be a valid alternative for ex-vivo skin absorption experiments.
