Summary: This study aimed to investigate the role of pravastatin in kanamycin-induced hearing loss in rats. The results demonstrated that pravastatin protected the cochlea from kanamycin-induced damage and exerted otoprotective effects by regulating autophagy and apoptosis.

Summary: Hearing loss is caused by a combination of genetic and environmental factors, with genetic factors accounting for 50% to 60% of cases. Mutations in mitochondrial DNA 12S rRNA, particularly the m.1555A>G mutation, are the most common molecular cause of nonsyndromic sensorineural hearing loss and aminoglycoside-induced deafness. The frequency of this mutation varies among different populations, possibly due to genetic differences among ancestors.

Summary: The study revealed that a single course of IV tobramycin can cause ototoxic hearing loss in some CF patients, emphasizing the need for routine ototoxicity monitoring and management in this clinical population. CF patients are suitable for clinical trials examining ototherapeutics in single IV tobramycin treatment episodes.

Summary: This pilot study successfully introduced a standardized tablet-based ototoxicity screening for pediatric CF patients. More research is needed to evaluate its impact and identify potential barriers. This method provides an accessible alternative to audiology testing and is crucial for early detection of medication-related toxicities in CF patients.

Summary: This study used a novel and scalable low-cost molecular technique called digital droplet polymerase chain reaction (ddPCR) to screen a large cohort of cystic fibrosis patients for the risk of aminoglycoside-induced hearing loss (AIHL) caused by genetic variants. The results demonstrate the utility of pretreatment screening using ddPCR in the cystic fibrosis population, which is cost-effective compared to other sequencing-based methods.

Summary: The human stria vascularis was examined in individuals who had received loop diuretics, aminoglycoside antibiotics, or a combination of the two. Structural changes were observed in the stria vascularis, which were similar to those seen in animal models. The study highlights the importance of using animal models to predict human ototoxicity.

Summary: Aminoglycosides, commonly used to treat infections in CF patients, are highly ototoxic. The study aimed to determine the incidence of ototoxicity after one course of once-daily IV tobramycin in CF patients. The results showed that cochleotoxic change was highly prevalent, and audiometric measures were more sensitive to ototoxic change than other assessment tools.

Summary: The consensus statement outlines four clinical recommendations for implementing routine and guideline adherent ototoxicity management in CF patients, including asking questions during routine CF case history, utilizing timely measures, establishing baselines and conducting evaluations, and repeating annual evaluations for patients with ototoxic antibiotic exposure. Efforts to implement an ototoxicity management program are crucial for improving identification of ototoxicity signs and symptoms, ensuring timely follow-up, and enabling informed decision-making for treatment modifications.

Summary: The study explores the psychological and emotional impact of hearing loss in adults with MDR-TB, as well as the social, economic, and vocational impact. It reveals that hearing loss has significant adverse effects on the lived experiences of MDR-TB patients, including stigma, discrimination, psychological distress, changes in family status and relationships, financial constraints, and social challenges. Unemployment poses a significant challenge, leading to economic instability and increased stress. Participants reported feelings of worthlessness, loss of identity, lack of motivation, embarrassment, and loss of independence.

Summary: The m.8296A>G variation in the mitochondrial tRNA(Lys) gene was found in a patient with sensorineural hearing impairment and other symptoms. Analysis of the mitochondrial proteome in this patient revealed different expressions of nuclear-encoded proteins involved in ATP synthesis and the Krebs cycle, suggesting pathogenicity of the variation with variable penetrance.
