Summary: The exposure to an acidic microenvironment initially reduced OSCC cells viability but led to an adaptation process. Acidic adapted cells acquired a mesenchymal-like phenotype with increased migration and motility indexes. Furthermore, tumoral extracellular acidity induced cellular stemness and increased chemo-and radioresistance in oral cancer cells.

Summary: MEX3A is an RNA-binding protein that promotes mRNA decay through binding to 30 untranslated regions. This study reveals that MEX3A is upregulated in clear cell renal cell carcinoma and its transcription is activated by ETS1. Silencing MEX3A reduces cell proliferation and induces G1/S cell-cycle arrest. Gene set enrichment analysis identified E2F targets as the central downstream pathways of MEX3A. Further assays demonstrated that MEX3A binds to the CDKN2B 30 untranslated region, promoting its mRNA degradation and leading to uncontrolled cell cycle in clear cell renal cell carcinoma.

Summary: Most human cells require anchorage for survival, and loss of cell-substrate adhesion can result in anoikis, a form of programmed cell death. Blebbing, the formation of small hemispherical plasma membrane protrusions, triggers the formation of signalling hubs that confer anoikis resistance by recruiting septin proteins as scaffolds for mutant NRAS and effectors. These signalling hubs activate pro-survival pathways, such as ERK and PI3K. Inhibition of blebs or septins in detached cells causes mislocalization of NRAS, reduced MAPK and PI3K activity, and cell death. Therefore, blebs play a crucial role in providing robust anoikis resistance.

Summary: Isopsoralen (IPRN), an active ingredient in Psoralea corylifolia Linn, demonstrated anti-inflammatory properties in rheumatoid arthritis (RA). Through inhibition of cytokine production, migration, invasion, and proangiogenic ability in RA fibroblast-like synoviocytes (FLSs), as well as reduction in paw thickness, arthritis score, bone damage, and serum inflammatory cytokine levels in mice with collagen-induced arthritis (CIA), IPRN showed promising antiarthritic effects in both in vitro and in vivo models. Additionally, the specific target of IPRN in RA FLSs was identified as macrophage migration inhibitory factor (MIF), suggesting therapeutic potential in RA treatment.
