Summary: This study presents a novel strategy for discovering CD147 glycosylation inhibitors through computer-aided screening, identifying compound 72 as the best candidate. Experimental results confirmed that compound 72 inhibited MMP production and cancer cell metastasis, and could promote E-cadherin expression by targeting CD147 to inhibit tumor migration. The structures of potential CD147 N-glycosylation inhibitors may guide future optimization efforts.

Summary: This study investigated the effect of estrogen on cancer-associated fibroblasts (CAFs) in gastric cancer and found that a subset of CAFs expressed estrogen receptor alpha (ER alpha). Estrogen could induce the expression of ER alpha in CAFs and promote the secretion of a paracrine factor CD147, which significantly enhanced cancer cell migration and invasion. High expression of CD147 was associated with poor prognosis in gastric cancer patients.

Summary: The study found that UGT1 from Polygonum tinctorium can catalyze N-, O-, or S-glycosylation with similar rates, and elucidated the molecular basis of N-, O-, and S-specificities using experimental mutagenesis and simulations. It also showed that the active site can be engineered to increase or favor one of the three glycosylation activities over another, leading to the design of more active and specific enzyme variants for glycoside production.

Summary: The study revealed that CD147 and HE4 co-immunoprecipitated and co-located in ovarian cancer tissues and cells, and were associated with disease progression and poor prognosis. HE4 was identified as an independent risk factor for CD147 in ovarian cancer tissues, and ANXA2 was also confirmed as a interacting protein of CD147 in ovarian cancer. Interaction between CD147 and HE4 can affect the invasion and metastasis of ovarian cancer.

Summary: The study revealed that reduced expression of Kidins220 was associated with tumorigenesis, metastasis, and overall survival in pancreatic cancer. Knockdown of Kidins220 promoted proliferation, colony formation, and tumorigenic capacity of pancreatic cancer cells, and regulated cell migration through the EGFR/AKT/ERK signaling pathway. Kidins220 also influenced cell invasion via MMP1 in pancreatic cancer cells.

Summary: This study examined the clinical value and therapeutic potential of ascitic autotaxin (ATX) in epithelial ovarian cancer (EOC). The results showed that elevated levels of ascitic ATX were associated with advanced stage, high grade, and a short disease-free period in EOC patients. Combining ascitic ATX and serum CA-125 levels improved the diagnostic accuracy for EOC, and functional studies revealed that ascitic ATX played a role in maintaining cancer stem cell-like characteristics and invadopodia formation in EOC.

Summary: Immunotherapy has achieved significant success in treating cancer, but resistance to this treatment often occurs due to the complex metabolic networks in the tumor microenvironment. Metabolic checkpoints play a crucial role in regulating T cell development, differentiation, and function, as well as coordinating metabolic competition between tumor cells and infiltrating T cells. Thus, targeting metabolic checkpoints may enhance the efficacy of immunotherapy.

Summary: Lung cancer is a common malignant tumor with a high mortality rate worldwide, especially non-small-cell lung cancer (NSCLC). Current treatment methods, such as radiotherapy, chemotherapy, targeted therapy, and surgery, often face challenges due to drug resistance and tumor invasion or metastasis. The ubiquitin-proteasome pathway (UPP) plays a critical role in tumor occurrence and development. Ubiquitin-specific proteases (USPs), as regulators of UPP, have the potential to be targeted for the treatment of NSCLC. This review focuses on the role of USPs in NSCLC and the potential of targeted drugs, PROTACs, and small-molecule inhibitors.
