Summary: Topical capsaicin has been shown to have acceptable time to resolution of symptoms and emergency department length of stay, making it an effective treatment option for symptomatic relief of Cannabinoid hyperemesis syndrome (CHS). Further randomized controlled trials are needed to determine its efficacy and efficiency in treating CHS across different care settings.

Summary: Neurostimulation appears to be a promising treatment option for chronic itch, with studies showing positive effects on various refractory itch conditions. Transcutaneous electrical nerve stimulation (TENS) was the most investigated modality, with atopic dermatitis being one of the most studied conditions in relation to neurostimulation. Further research is needed to better understand the mechanisms of action of neurostimulation in different pruritic conditions.

Summary: This review evaluated the therapeutic effects and safety of Chinese herbal medicine (CHM) for the treatment of chronic pruritus. Analysis of 24 randomized controlled trials showed that CHM treatment was more effective than Western medicine treatment in relieving symptoms. However, high-quality studies are needed to further validate these findings.

Summary: The topical administration of medicines for retinal degenerative diseases is limited by the eye's barriers, but NSAIDs have shown potential for effective treatment. Other drug classes may require fine-tuning of pharmacokinetic profiles or advanced formulation strategies for improved delivery to the retina. Ultimately, developing more effective ways of delivering drugs to the posterior eye will enhance clinical efficacy.

Summary: Chronic kidney disease-associated pruritus (CKD-aP) significantly reduces the quality of life for patients with end-stage renal disease. The specific cause of CKD-aP is not fully understood, but an imbalance in the opioid pathways, particularly the downregulation of the kappa-opioid receptor, may contribute to itching sensation. Difelikefalin, a selective kappa-opioid receptor agonist, has recently been approved as the first drug for treating pruritus associated with chronic kidney disease (CKD) in adult hemodialysis patients. A systematic review of clinical trials found that difelikefalin effectively reduces pruritus and improves quality of life in CKD patients. The most common side effects observed were mild, including nausea, vomiting, dizziness, and diarrhea. Due to its proven efficacy and good safety profile, difelikefalin shows promise as a treatment for pruritus in patients with chronic kidney disease.

Summary: Alzheimer's Disease (AD) is a progressive neurodegenerative disorder characterized by cognitive impairment and specific neuropathological changes. Capsaicin, a compound found in chili peppers, has shown anti-inflammatory, antioxidant, and potential neuroprotective properties. Studies have suggested that capsaicin intake may improve cognitive function and reduce tau pathology in AD. This systematic review discusses the potential benefits of capsaicin in improving AD pathology and symptoms based on studies using animal models and cell cultures.
