Summary: We aimed to propose a cell cycle-related multi/mRNA signature (CCS) for prognosis prediction and uncover new tumor-driver genes for hepatocellular carcinoma (HCC). The CCS was constructed using statistical analysis and Cox regression analyses. Predictive capacity and clinical practicality of the signature were evaluated and validated. TICRR was identified as a tumor-driver gene for HCC through functional experiments.

Summary: This study explored the expression and function of 40S ribosomal protein S24 (RPS24) in hepatocellular carcinoma (HCC). Results showed that RPS24 was elevated in HCC samples and related to poor prognosis. Furthermore, RPS24 overexpression or promoter methylation correlated with unfavorable prognosis in HCC patients. High expression of RPS24 was associated with DNA replication, cell cycle, immune infiltration, and immunotherapy response. Experimental validation demonstrated that silencing of RPS24 suppressed HCC cell growth and tumor proliferation. Therefore, RPS24 may serve as a potential adverse biomarker for HCC prognosis, acting through cell proliferation facilitation and the formation of an immunosuppressive microenvironment, and targeting RPS24 could be a promising therapeutic option for HCC management.

Summary: Hepatocellular carcinoma (HCC) is a global health challenge with increasing incidence worldwide. The circadian clock has been shown to play a key role in hepatocarcinogenesis, but the cellular and molecular mechanisms underlying the HCC-clock crosstalk are unknown. In this study, we found that HCC cells rely on the circadian clock transcription factors BMAL1 and CLOCK for cell growth, and down-regulation of these factors induces apoptosis and cell cycle arrest. Mechanistically, inhibiting BMAL1/CLOCK dysregulates cell cycle regulators Wee1 and p21, leading to tumor cell death. Our findings provide insights into the cellular impact of clock proteins in HCC oncogenesis and suggest a potential therapeutic approach based on modulation of the circadian clock.
