Summary: Multiple studies have shown that exposure to micro- or nano-plastics can damage the viability and function of macrophages in intestinal tissue, possibly due to cell apoptosis and inflammation. In this study, the cytotoxicity and pro-inflammatory effects of 80 nm polystyrene nano-plastics (PS-NP) and 3 μm polystyrene micro-plastics (PS-MP) on mouse macrophages were investigated. The results demonstrated that both PS-NP and PS-MP induced cell apoptosis, and PS-NP had a more potent inflammatory effect by enhancing the secretion of inflammatory cytokines. Mechanistically, PS-NP and PS-MP were found to cause cytotoxicity and inflammation by producing reactive oxygen species and nitric oxide in the cells. Overall, this study highlighted the cytotoxicity and pro-inflammatory effects of PS-NP and PS-MP on macrophages, which could lead to intestinal inflammation, with PS-NP showing greater impacts.

Summary: This study investigates the role of hypoxia-inducible factor (HIF)-1 alpha/heme oxygenase (HO)-1 in hippocampal neuron ferroptosis during epilepsy. The results suggest that there are metabolic differences between epilepsy patients and healthy individuals, with increased oxidative stress, heightened bilirubin, and augmented iron metabolism in epileptic blood. The active HIF-1 pathway in epileptic pathogenesis and perturbations in HIF-1 alpha signaling are reinforced by the analysis. The study implicates that HIF-1 alpha/HO-1 potentially regulates hippocampal neuronal ferroptosis, iron metabolism, and oxidative stress, thereby promoting the propagation of epilepsy.

Summary: A series of diclofenac hybrid molecules were synthesized and evaluated for their ability to inhibit NO in LPS-induced RAW 264.7 macrophage cells. Among them, compound 1 showed the highest NO-inhibitory ability (approximately 66%) with no significant cytotoxicity. Compound 1 also exhibited superior NF-KB-inhibitory ability compared to diclofenac by activating the Nrf2/HO-1 signaling pathway. Additionally, compound 1 demonstrated promising anti-inflammatory effects in a DSS-induced mice model of colitis and showed lower ulcerogenic potential compared to diclofenac in rats. The diclofenac-eugenol hybrid (compound 1) has the potential to serve as a novel anti-inflammatory agent due to its ability to inhibit the NF-KB signaling pathway and activate HO-1 expression, both in vitro and in vivo, without toxicity.

Summary: The activation of the inflammasome plays a critical role in the pathogenesis of systemic lupus erythematosus (SLE), and mesenchymal stem cells (MSC) have been shown to alleviate SLE by suppressing inflammasome activation. This study found that the NLRP3 inflammasome was activated in macrophages from SLE patients and mice, and its activation correlated with disease activity. After MSC transplantation, the severity of SLE was reduced, and NLRP3 inflammasome activation was inhibited.
