Azathioprine-induced toxoplasma gondii infection in a patient with Crohn's disease with NUDT15 variation A case report

Summary: This case report highlights a Crohn's disease patient who developed toxoplasma gondii infection after treatment with azathioprine, with a genetic variation in NUDT15 but normal TPMP gene. NUDT15 variation may contribute to reduced tolerance to azathioprine and personalized therapy based on genetic testing may help reduce the risk of adverse events.

Summary: The study found that in patients with ulcerative colitis (UC) and Crohn's disease (CD), the gut microbiome was more similar to the oral microbiome, indicating increased ectopic gut colonization by oral bacteria in IBD patients. Incipient periodontitis did not significantly affect the clinical outcomes of UC and CD patients, but early periodontitis may be associated with worse clinical symptoms in some CD patients.

Summary: The development of Crohn's disease is characterized by a breakdown in the immune-bacterial communication, leading to a failure in mucosal defense mechanisms. This can result in a compromised epithelial barrier and dysregulated immune responses, ultimately causing chronic inflammation in the intestinal tissue. Understanding these cellular and molecular mechanisms may lead to targeted therapeutic approaches for treating CD.

Efficacy of Combined Initial Treatment of Methotrexate with Infliximab in Pediatric Crohn's Disease: A Pilot Study

Summary: Oxidative stress is important in the pathogenesis of inflammatory bowel diseases (IBDs). Superoxide dismutase (SOD) defenses are weakened in IBD patients. A Mn(II) complex SOD mimic (Mn1) based on an open-chain diaminoethane ligand has antioxidant and anti-inflammatory effects. Metal exchanges occur between the manganese center and metal ions in the biological environment. New Mn(II) complexes with cyclohexyl and/or propyl groups show improved SOD activity and kinetic inertness in metal ion exchange processes. They provide anti-inflammatory and antioxidant effects in cells at lower doses than Mn1.

Summary: The study found that the luxR gene from Bacteroides dorei was under-expressed in IBD patients, while one luxR receptor gene from Bacteroides fragilis appeared to be over-expressed. Targeting LuxR receptors of bacterial quorum sensing might represent a new approach to modulate the gut microbiota in IBD.

Urbanization associates with restricted gut microbiome diversity and delayed maturation in infants

Summary: This study compared the gut microbiomes of mother and infant pairs from rural and urban areas of Senegal. The results showed that urban mothers had different gut microbiome compositions than rural mothers, while urban infants had delayed gut microbiome maturation and higher susceptibility to infectious diseases. These findings suggest that industrialization is associated with gut microbiome alterations and may be linked to disease.
