Gapdh Gene Expression Is Modulated by Inflammatory Arthritis and Is not Suitable for qPCR Normalization

Gene expression studies are fundamental for the understanding of complex diseases, providing new insights into the pathogenic process and new tools for diagnostic and patient stratification. Gene profiling studies by real-time PCR require the use of reference genes for normalization and an appropriate validation is essential for accurate results. We performed a comprehensive assessment of six common housekeeping genes in the K/BxN serum-induced arthritis model in mice. Classical statistics and NormFinder analyses pointed out Gapdh as the less stable and therefore unsuitable as a reference control. Gapdh was considerably down-regulated in arthritic joints and therefore produced an overestimation of transcriptional changes. Hptr, B2m, and Rpl13a showed the most constant expression. Collectively our data advise against the use of Gapdh in gene expression studies in the acute phase of the K/BxN model and adds a cautionary note on the need to validate the reference genes for reliable, comparable, and reproducible results.

Senescence under appraisal: hopes and challenges revisited

Summary: Cellular senescence, defined as an irreversible cell cycle arrest with increased cellular growth and metabolic activity, has become a focus of biomedical research. The dual nature of senescence, beneficial/detrimental, is linked to contextual aspects, with the pro-resolving model suggesting that pathology arises from incomplete senescence program. Therapeutic approaches based on senescence include induction, prevention, and clearance of senescent cells, as well as the use of senolytic and senomorphic drugs. Challenges and gaps in research need to be addressed to promote the clinical success of senescence-based therapies.

Extracellular vesicles from monocyte/platelet aggregates modulate human atherosclerotic plaque reactivity

Summary: Extracellular vesicles released by mixed aggregates of monocytes and platelets have pro-inflammatory effects on endothelial cells and atherosclerotic plaques, and modulating platelet activation can alter the composition and actions of these vesicles, potentially providing therapeutic avenues for atherosclerosis treatment. Patients with coronary artery disease requiring intervention show elevated levels of specific EV subsets in plasma.

Summary: Regulatory T-cells play a central role in maintaining homeostasis and resolving inflammation. ALOX15 was found to regulate key aspects of T-reg biology, with its inhibition or deletion leading to altered gene expression and metabolism in T-regs. Incubation with ALOX15-derived pro-resolving mediators rescued the expression of key regulatory genes in T-regs with inhibited ALOX15 activity.
