To study the specific role of transmembrane tumor necrosis factor (tmTNF) in protective and pathological responses against the gastrointestinal helminth Trichinella spiralis, we compared the immune responses of TNF-alpha/lymphotoxin alpha (LT alpha)(-/-) mice expressing noncleavable transgenic tmTNF to those of TNF-alpha/LT alpha(-/-) and wild-type mice. The susceptibility of TNF-alpha/LT alpha(-/-) mice to T. spiralis infection was associated with impaired induction of a protective Th2 response and the lack of mucosal mastocytosis. Although tmTNF-expressing transgenic (tmTNF-tg) mice also had a reduced Th2 response, the mast cell response was greater than that observed in TNF-alpha/LT alpha(-/-) mice and was sufficient to induce the expulsion of the parasite. T. spiralis infection of tmTNF-tg mice resulted in significant intestinal pathology characterized by villus atrophy and crypt hyperplasia comparable to that induced following the infection of wild-type mice, while pathology in TNF-alpha/LT alpha(-/-) mice was significantly reduced. Our data thus indicate a role for tmTNF in host defense against gastrointestinal helminths and in the accompanying enteropathy. Furthermore, they also demonstrate that TNF-alpha is required for the induction of Th2 immune responses related to infection with gastrointestinal helminth parasites.

Summary: The study developed ZIKV subunit and virus-like particle (VLP)-based vaccines, which induced antibodies with a higher capacity to neutralize virus infection and protected animals from lethal challenge with both the African and Asian lineages of ZIKV. The engineered immunogens reduced the potential to induce antibody-dependent enhancement (ADE) of infection and showed promising efficacy in comparison with the wild-type derivatives.

Epithelial-to-mesenchymal transition contributes to the downregulation of progesterone receptor expression in endometriosis lesions

Summary: This study found a negative correlation between low expression of progesterone receptors (PR) and epithelial-to-mesenchymal transition (EMT) in endometriosis lesions. In both in vitro and in vivo experiments, EMT leads to downregulation of PR gene expression by upregulating EMT transcription factors like SNAI1 and SNAI2, potentially resulting in progesterone resistance.

Molecular and Histological Profiling Reveals an Innate-Shaped Immune Microenvironment in Solitary Juvenile Polyps

Summary: Solitary juvenile polyps (JP) exhibit a benign disease course with low recurrence rate but with signs of intestinal inflammation. Study results showed predominant accumulation of neutrophils and eosinophils in JP, with increased levels of CXCL8, CXCL5, and CCL11 transcripts suggesting enhanced recruitment of these cell types. Proinflammatory cytokines and inflammation-amplifying receptors were higher in JP compared with IBD, indicating a different immune response mechanism.

Streptococcus pneumoniae Rapidly Translocate from the Nasopharynx through the Cribriform Plate to Invade the Outer Meninges

Teerawit Audshasai, Jonathan A. Coles, Stavros Panagiotou, Shadia Khandaker, Hannah E. Scales, Morten Kjos, Murielle Baltazar, Julie Vignau, James M. Brewer, Aras Kadioglu, Marie Yang

Summary: Using two-photon imaging, we demonstrate that pneumococci and microspheres can translocate from the nasal cavity to the meninges of mice within minutes of inhaled delivery. This suggests the existence of an inward flow of fluid connecting the nasopharynx to the meninges. Our findings provide new insights into the mechanisms of pathogens invading the central nervous system and have implications for drug delivery and air pollution-related brain damage.
