Objective : To assess the anticataleptic and antiepileptic activity of leaves of Mucuna pruriens in albino rats. Materials and Methods : Haloperidol-induced catalepsy (HIC), maximum electro-shock (MES) method, pilocarpine-induced Status epilepticus (PISE) and single-dose effect of M. pruriens were employed. Results : M. pruriens (100 mg/kg) had significant anticataleptic and antiepileptic activity in HIC, MES, and PISE. Conclusions : M. pruriens extract has the potential to be an anticataleptic and antiepileptic drug. Dopamine and 5-HT may have a role in such activity.

Coumarin derivatives as promising antibacterial agent(s)

Chita Ranjan Sahoo, Jyotirmaya Sahoo, Monalisa Mahapatra, Debananda Lenka, Pratap Kumar Sahu, Budheswar Dehury, Rabindra Nath Padhy, Sudhir Kumar Paidesetty

Summary: Bacterial infections are significant health threats worldwide, with antibiotic resistance posing a challenge. The development of new potential drug candidates is necessary to combat multidrug-resistant bacteria. Coumarin and its derivatives have been reported to have broad inhibitory properties, with substituents at positions C-3 and C-4 shown to be coveted for the development of newer antibacterial agents.

Captopril is more effective than Perindopril against aluminium chloride induced amyloidogenesis and AD like pathology

Debashish Mohapatra, Srikant Kanungo, Sweta Priyadarshini Pradhan, Susmita Jena, Shakti Ketan Prusty, Pratap Kumar Sahu

Summary: In this study, the effects of Captopril and Perindopril on cognitive dysfunction and amyloidogenesis induced by aluminium chloride were evaluated and compared. The results showed that both Captopril and Perindopril prevented and improved cognitive dysfunction, reduced amyloid-beta content, and improved antioxidant status. They also reversed the histopathological changes in the hippocampus region. Captopril was found to be more effective than Perindopril.

Photocatalysis of environmental organic pollutants and antioxidant activity of flavonoid conjugated gold nanoparticles

Sweta Priyadarshini Pradhan, Sunsita Swain, Nishigandha Sa, Satya Narayan Pilla, Anindita Behera, Pratap Kumar Sahu, Sudam Chandra Si

Summary: Nanomaterials have unique properties that make them potentially useful in biomedical applications and environmental pollutant management. This research focuses on isolating hesperidin from orange peel and preparing hesperidin gold nanoparticles using a chemical reduction method. The gold nanoparticles have high substrate specificity and can be excited by visible light. They show excellent efficacy in managing and removing organic pollutants. The study also demonstrates that hesperidin gold nanoparticles have significant antioxidant and photocatalytic activity, making them valuable in disease management and wastewater treatment.

Mechanistic study of gold nanoparticles of Vildagliptin and Vitamin E in diabetic cognitive impairment

Sweta Priyadarshini Pradhan, P. Tejaswani, Nishigandha Sa, Anindita Behera, Rajesh Kumar Sahoo, Pratap Kumar Sahu

Summary: The present study investigates the efficacy of antidiabetic drug VLD and antioxidant VE against STZ-induced diabetic cognitive impairment. To enhance the efficacy of VLD, gold nanoparticles are conjugated with VLD for targeted brain delivery. The treatment with drug-conjugated Au NPs improves cognitive functions and shows neuroprotective action against diabetes-induced cognitive impairment.

New insights toward molecular and nanotechnological approaches to antidiabetic agents for Alzheimer's disease

Sweta Priyadarshini Pradhan, Pratap Kumar Sahu, Anindita Behera

Summary: Alzheimer's disease (AD) and type-II diabetes mellitus (DM) are mutually related chronic neurodegenerative disorders. Chronic DM increases the risk for AD due to insulin resistance and cognitive dysfunction. Repurposing of antidiabetic drugs and nanotechnological interventions show promise in the prevention and treatment of AD. This review explores the cellular and molecular pathophysiologies of AD and type-II DM, the use of in vivo DM models in AD studies, and the potential of antidiabetic drugs and nanodelivery systems for AD treatment.

Ameliorative and Neuroprotective Effect of Core-Shell Type Se@Au Conjugated Hesperidin Nanoparticles in Diabetes-Induced Cognitive Impairment

Anindita Behera, Sweta Priyadarshini Pradhan, P. Tejaswani, Nishigandha Sa, Swagata Pattnaik, Pratap Kumar Sahu

Summary: Diabetes mellitus leads to cognitive dysfunction due to insulin deficiency. Hesperidin, a bioflavonoid, has poor solubility and a short half-life. To overcome these limitations, hesperidin was conjugated with gold, selenium, and core-shell bimetallic nanoparticles. The synthesized nanoparticles showed neuroprotective effects in a rat model of cognitive dysfunction induced by streptozotocin. Among the nanoparticles, the bimetallic Se@Au-HSP NPs exhibited the strongest neuroprotective action. Hesperidin-conjugated bimetallic nanoparticles could be a potential alternative for neurodegenerative diseases.
