Summary: Although the identification of genes involved in NDDs has improved our understanding of their causes, obstacles still remain in developing therapies for ID and other NDDs due to clinical and genetic heterogeneity, rarity of recurrent pathogenic variants, and comorbidity with psychiatric traits. A broad range of clinical symptoms in some NDDs is caused by a large intragenic mutational landscape, with different DNA variations affecting protein functions and downstream processes. This review focuses on understanding the molecular changes of group I PAKs and their contribution to the clinical spectrum of NDDs. By identifying differences in PAK structure, regulation, and expression, we can uncover the mechanisms underlying mutation pathogenicity and develop personalized therapeutic approaches.

Summary: This case report describes a patient with LETM associated with Dengue virus infection who responded well to treatment. A systematic review of published cases found three additional cases meeting criteria for NMOSD. The study suggests that early appropriate immunotherapy is crucial for patients suspected of having NMOSD.

Summary: BIA-ALCL, though declining clinically, is still an important topic in breast implant safety discussions. Plastic surgeons need to understand the disease progression and classification, use high-level surgical techniques to reduce bacterial load, and practice evidence-based surgery with vigilant postoperative follow-up care to improve outcomes.

Summary: This article reports on two completely asymptomatic mothers of FARAD patients, emphasizing that FARAD is an important differential diagnosis of genetically determined congenital neuromuscular disorders even in asymptomatic mothers. The article also suggests that IVIG therapy during pregnancy has the potential to improve the outcome of the children.
