Summary: Ulcerative colitis increases the risk of colorectal cancer, and this study examined the genetic and epigenetic profiles of colitis-associated colorectal carcinomas (CA-CRC). Subgroups within CA-CRC were identified with different mutational loads and signatures, which may guide clinical management and therapy options. Lynch syndrome tumors showed a higher degree of hypermutability compared to CA-CRC subgroups.

Summary: This study identifies and characterizes polymorphic human leukocyte antigen (HLA)-class II genes, MTCO3P1 pseudogene alleles, and indels of transposable elements linked to specific HLA-class II alleles. The research also reveals unique crossover sites between SNP-poor and SNP-rich genomic segments, with significant haplotype shuffling near putative recombination hotspots.

Summary: The leaf extract of Plantago major exhibits anti-inflammatory effects and protects against mucosal damage in rats with ulcerative colitis, especially when administered in high doses. The seed extract of P. major has minimal protective effects on ulcerative colitis.

Summary: This study investigated the variability of the immune response-related gene BoLA-DQA1 in cattle from Nigeria and New Zealand. Ten alleles were identified using a PCR-SSCP approach, with two novel alleles reported for the first time through DNA sequencing. Overall, 12.60% of nucleotide positions analyzed were found to be variable.

Summary: The study analyzed MHC class II and microsatellite diversity in 16 guppy populations from Trinidad and Tobago, showing high within-population MHC diversity with limited allele sharing both within and between islands. Phylogenetic analyses suggested island-specific allelic lineages, and the generation of new alleles at a detectably high frequency. Balancing selection was proposed as the general process behind spatial variation in MHC allele frequencies in this system, consistent with Red Queen processes of host-parasite coevolution.

Summary: In this study, we identified and characterized 11 polymorphic microsatellite markers suitable for routine testing in dromedaries and Bactrian camels. These markers can be used to study the adaptive genetic diversity of the MHC region in Old World camels.

Summary: Biologic agents and oral small molecules are the main treatments for IBD, but there is a need for new agents with novel mechanisms of action. miRNAs may be potential therapeutic targets for IBD. Obezafimod, a first-in-class oral small molecule, has shown promise in phase II trials for UC patients.

Summary: Major Histocompatibility Complex (MHC) Class II deficiency is a rare autosomal recessive primary immunodeficiency caused by mutations in regulatory genes of MHC Class II proteins. This case report describes a 9-month-old girl presenting with repeated pneumonia, lymphocytopenia, and absence of HLA-DR expression on B cells, which led to the diagnosis of MHC Class II deficiency. Genetic analysis identified a homozygous mutation in the RFX-5 gene. Despite the patient's unfortunate outcome, the importance of prenatal diagnosis was emphasized to the parents.

Summary: The study revealed that most patients in the UC OCTAVE program had a low baseline 10-year ASCVD risk. MACE occurred more frequently in patients with prior ASCVD and higher baseline CV risk, suggesting the importance of assessing individual CV risk in clinical practice.

Summary: This study aimed to investigate the incidence and factors associated with the development of postoperative perianal fistula in patients undergoing ileoanal pouch anastomosis for ulcerative colitis. The study found that younger age at disease onset, lower age at surgery, and postoperative diagnosis of Crohn's disease or indeterminate colitis were correlated with perianal fistulas.
