In research aimed at improving human health care, animal studies still play a crucial role, despite political and scientific efforts to reduce preclinical experimentation in laboratory animals. In animal studies, the results and their interpretation are not always straightforward, as no single study is executed perfectly in all steps. There are several possible sources of bias, and many animal studies are replicates of studies conducted previously. Use of meta-analysis to combine the results of studies may lead to more reliable conclusions and a reduction of unnecessary duplication of animal studies. In addition, due to the more exploratory nature of animal studies as compared to clinical trials, meta-analyses of animal studies have greater potential in exploring possible sources of heterogeneity. There is an abundance of literature on how to perform meta-analyses on clinical data. Animal studies, however, differ from clinical studies in some aspects, such as the diversity of animal species studied, experimental design, and study characteristics. In this paper, we will discuss the main principles and practices for meta-analyses of experimental animal studies.

First step in implementation of opportunistic salpingectomy for prevention of ovarian cancer: Current care and its determinants

Malou E. Gelderblom, Veerle Jaspers, Reinier P. Akkermans, Brigitte Slangen, Angele L. M. Oei, Alexandra A. van Ginkel, Huy Ngo, Joanna IntHout, Rosella P. M. G. Hermens, Joanne A. de Hullu, Jurgen M. J. Piek

Summary: This study aimed to determine the implementation of counseling and performance of opportunistic salpingectomy (OS) between 2015 and 2018, and its patient, surgical, physician, and hospital characteristics. The results showed that the proportion of counseling and performance increased over the years, but there were still variations. Therefore, an implementation strategy tailored to associated determinants is recommended.

Menon, Ali Mobasheri, Nikki Osborne, Merel Ritskes-Hoitinga, Bettina Ryll, Elmar Schmitt, Anton Ussi, Antonio L. Andreu, Emmet McCormack

Summary: Personalized medicine aims to provide tailor-made prevention and treatment strategies for specific groups, but there are challenges in clinical relevance and model validity. Researchers have developed a set of recommendations to improve the robustness of preclinical methods in translational research for personalized medicine through literature review, expert discussions, and consensus meetings.

Contraceptives and cancer risks in BRCA1/2 pathogenic variant carriers: a systematic review and meta-analysis

Majke H. D. van Bommel, Joanna IntHout, Guus Veldmate, C. Marleen Kets, Joanne A. de Hullu, Anne M. van Altena, Marline G. Harmsen

Summary: The use of oral contraceptive pills may increase the risk of breast cancer but decrease the risk of ovarian cancer in BRCA1/2-PV carriers. Tubal ligation also offers protection against ovarian cancer. Counselling for BRCA1/2-PV carriers should be personalized, taking into account genetic and non-genetic factors as well as patient preferences.

Subjective outcomes 12 years after transvaginal mesh versus native tissue repair in women with recurrent pelvic organ prolapse; a randomized controlled trial

Kirsten B. Kluivers, Metteke Kamping, Alfredo L. Milani, Joanna IntHout, Mariella I. Withagen

Summary: This study is an extended follow-up study after 12 years, focusing on the subjective outcomes of women who underwent surgery for recurrent pelvic organ prolapse. The results showed no difference in subjective outcomes between the groups, but there was a statistically significant higher number of women who required further operations in the mesh group.

The effectiveness of transabdominal cerclage placement via laparoscopy or laparotomy: a systematic review and meta-analysis

Cecile C. Hulshoff, Aniek Hofstede, Joanna Inthout, Ralph R. Scholten, Marc E. A. Spaanderman, Hanna Wollaars, Joris van Drongelen

Summary: This systematic review and meta-analysis compared the outcomes of laparoscopic and open laparotomy abdominal cerclage approaches in women with cervical insufficiency. The results showed that both methods had similar survival rates and gestational age at delivery. However, there were some differences in perioperative care, surgical complications, and complications during pregnancy.

Melissa J. J. van Velthoven, Aksel N. Gudde, Frederique Struijs, Egbert Oosterwijk, Jan-Paul Roovers, Zeliha Guler, Carlijn R. Hooijmans, Paul H. J. Kouwer

Summary: Growth factors, especially basic fibroblast growth factor, promote vaginal wound healing in vitro, but the effects are less prominent in vivo. More research is needed to improve surgical outcomes for pelvic organ prolapse, including improved preclinical models.

The 3Ranker: An AI-based Algorithm for Finding Non-animal Alternative Methods

Niels van Beuningen, Sinne Alkema, Nils Hijlkema, Brun Ulfhake, Rafael Frias, Merel Ritskes-Hoitinga, Wynand Alkema

Summary: This paper introduces a fast algorithm called 3Ranker for finding non-animal alternative methods in biomedical research. By utilizing machine learning and expert-curated data, the algorithm is able to accurately identify potential alternatives to animal use in a large number of articles. The algorithm performs well with Area Under the Curve (AUC) values ranging from 0.85-0.95 and demonstrates differences in the early implementation of the Three Rs principles across different research domains.

Ilaria Viozzi, Maroeska M. Rovers, Christiaan G. Overduin, Mark ter Laan

Summary: Introduction: The application of stereotactic laser ablation (SLA) for intra-cranial tumours has been growing, but comparative trials are lacking. This study investigated the familiarity with SLA and opinion on potential indications of European neurosurgeons. Results showed that most respondents considered SLA as a treatment option for recurrent glioblastoma, recurrent metastases, and newly diagnosed deep-seated glioblastoma.

The accuracy of navigated versus freehand curettage in bone tumors: a cadaveric model study

Thomas R. F. van Steenbergen, Han Nijsink, Thomas G. E. Eggen, Dennis Janssen, Maroeska M. Rovers, Ingrid C. M. van der Geest, J. J. Futterer

Summary: In this exploratory study on 20 cadaveric bone tumor models, navigated curettage was not found to be more accurate than freehand curettage. However, the presence of remaining tumor indicates that there is still room for improvement in curettage procedures. The novice user was less accurate with navigation compared to freehand, which could be attributed to the learning curve. Furthermore, the expert user employed a different surgical approach, focusing more on complete tumor removal rather than preserving the surrounding bone.
