Neuroprotection against 6-OHDA-induced oxidative stress and apoptosis in SH-SY5Y cells by 5,7-Dihydroxychromone: Activation of the Nrf2/ARE pathway

Aims: The aim of this study was to prove the neuroprotective effect of 5,7-Dihydroxychromone (DHC) through the Nrf2/ARE signaling pathway. To elucidate the mechanism, we investigated whether 6-hydroxydopamine (6-OHDA)-induced neurotoxicity in SH-SY5Y cells could be attenuated by DHC via activating the Nr12/ARE signal and whether DHC could down-regulate 6-OHDA-induced excessive ROS generation Main methods: To evaluate the neuroprotective effect of DHC against 6-OHDA-induced apoptosis, FACS analysis was performed using PI staining. The inhibitory effect of DHC against 6-OHDA-induced ROS generation was evaluated by DCFH-DA staining assay. Additionally, translocation of Nrf2 to the nucleus and increased Nrf2/ARE binding activity, which subsequently resulted in the up-regulation of the Nrf2-dependent antioxidant gene expressions including HO-1, NQO1, and GCLc, were evaluated by Western blotting and EMSA. Key findings: Pre-treatment of DHC, one of the constituents of Cudrania tricuspidata, significantly protects 6-OHDA-induced neuronal cell death and ROS generation. Also, DHC inhibited the expression of activated caspase-3 and caspase-9 and cleaved PARP in 6-OHDA-induced SH-SY5Y cells. DHC induced the translocation of Nrf2 to the nucleus and increased Nrf2/ARE binding activity which results in the up-regulation of the expression of Nrf2-dependent antioxidant genes, including HO-1, NQO1, and GCLc. The addition of Nrf2 siRNA abolished the neuroprotective effect of DHC against 6-OHDA-induced neurotoxicity and the expression of Nrf2-mediated antioxidant genes. Significance: Activation of Nrf2/ARE signal by DHC exerted neuroprotective effects against 6-OHDA-induced oxidative stress and apoptosis. This finding will give an insight that activating Nrf2/ARE signal could be a new potential therapeutic strategy for neurodegenerative disease. (C) 2015 Elsevier Inc. All rights reserved.

Natural photosensitizers from Tripterygium wilfordii and their antimicrobial photodynamic therapeutic effects in a Caenorhabditis elegans model

Summary: The extract and photosensitizer-enriched fraction of Tripterygium wilfordii showed effective antimicrobial photodynamic treatment (APDT) effects, particularly against Staphylococcus aureus, methicillin-resistant Staphylococcus aureus (MRSA), and Candida albicans. Additionally, the photosensitizers were able to induce intracellular ROS production and showed strong APDT activity against skin pathogens like Staphylococcus epidermidis and Streptococcus pyogenes. Overall, the use of TWE and TW-F5 in APDT was found to be successful in controlling pathogenic bacteria.

Summary: Eight new naphtho [1,2-c]furan derivatives and six known analogues were isolated from the culture medium of Basidioradulum radula, with their structures identified through spectroscopic analysis and absolute configurations resolved using various methods. Compounds 7 and 14 exhibited anti-cancer activity against human prostate cancer cells, but had no effect on the proliferation of splenocytes from immunosuppressed mice.

This study evaluated the inhibitory effects of obtusifolin and its analogs on CYP1A enzymes and investigated their inhibitory mechanism and chemopreventive effects. The results showed that obtusifolin selectively inhibited CYP1A2 activity and exhibited potential chemopreventive effects.

Summary: In this study, 13 compounds, including three new monoterpenoids, were isolated from the leaves of Actinidia polygama. The structures of these compounds were characterized and their potential bioactivities were investigated. Two compounds, 3-O-coumaroylmaslinic acid and jacoumaric acid, showed insulin secretion activity and alpha-glucosidase inhibitory activity. Furthermore, they increased the expression of insulin-related proteins and may be developed as potential antidiabetic agents.

Collinsella aerofaciens Produces a pH-Responsive Lipid Immunogen

Summary: Some members of the human gut microbiota have significant impact on the host's physiology, health, and therapeutic responses, but the specific molecules and mechanisms responsible are not well understood. As part of a research project, the metabolome of Collinsella aerofaciens, a commonly studied actinobacterium, was analyzed to identify immune-regulating compounds produced by gut microbes. The active compound, CaLGL-1, a lysoglycoglycerolipid, was identified and its production was found to be dependent on TLR2 signaling.

Summary: Gastrointestinal stromal tumors (GISTs) frequently show KIT mutations, leading to overexpression and abnormal localization of mutant KIT (MT-KIT). BLZF1 was identified as a novel binding partner of MT-KIT, anchoring it to the Golgi complex. The sustained activation of ATF6, a member of the unfolded protein response (UPR) family, helps in MT-KIT folding and relieves endoplasmic reticulum (ER) stress. The activation of ATF6 is frequently observed in GIST patients and is significantly associated with poorer relapse-free survival.

Antimicrobial photodynamic therapy with Ligularia fischeri against methicillin-resistant Staphylococcus aureus infection in Caenorhabditis elegans model

The findings suggest that this PE could be a potential treatment for AD.
