The sex ratio of offspring is associated with the mothers' age at menarche

BACKGROUND: Early menarcheal age is a risk factor for breast and ovarian cancers and is also associated with an increased spontaneous abortion rate. The aim of this study was to investigate whether there is a link between early menarcheal age and the offspring sex ratio. METHODS: We recorded the sex of 21 208 live born infants, all singletons, born to 10 847 premenopausal women (mean attending age: 37.5 +/- 7.2 years, range 22-54) who attended our clinics for obstetrical and gynaecological assessment. We calculated the sex ratio of newborn infants in relation to the mothers' age of menarche (from 9 to 18 years) and to the number of infants per woman (i.e. fertility index). RESULTS: A low offspring sex ratio (males/females) of 0.800 was observed in mothers who entered menarche at the age of 9 years; the odds ratio (OR) (95% confidence intervals) compared with those of control group with menarche at age 14 was 0.72 (0.29-1.79). The sex ratio remained low, until the group of mothers who entered menarche at 12 years of age [1.009; OR: 0.90 (0.85-0.95)]. The highest sex ratio was observed in mothers with a menarcheal age of 14 years (1.118; OR: 1.00 control) and it gradually decreased until the menarcheal age of 17 years [1.000; OR: 0.89 (0.15-5.26)]. The fertility index was also low in women with menarche at the age of 9 to 11 years. CONCLUSIONS: Women entering menarche outside the normal range, especially those with earlier menarche, may have an increased chance of producing female offspring.

Summary: The study found that adding 70 or 250 IU/L FSH to the IVM medium improved oocyte nuclear maturation by regulating gene expression in cumulus cells, suggesting a threshold for FSH concentration during IVM of human oocytes from small follicles. Moreover, FSH functions in vitro by downregulating FSHR and upregulating LHCGR in cumulus cells, which facilitates the maturation of immature oocytes.

Summary: A study identified 290 genetic determinants of ovarian aging, associated with DNA damage response processes and loss-of-function variants in key DDR-associated genes. Experimental models showed that these DDR processes act across the life-course to shape ovarian reserve and its rate of depletion. Extending reproductive life in women may improve bone health and reduce the risk of type 2 diabetes, but it could increase the risk of hormone-sensitive cancers.

Histidine buffered media maintains pH stabile during cooled transportation of human ovarian tissue

Summary: This study aimed to investigate the stability of pH when transporting ovarian tissue in media buffered with HEPES or histidine, and the impact of transport media choice on in vitro maturation rate of oocytes collected in connection with ovarian tissue cryopreservation. The results showed that the pH in histidine buffered media is closer to physiological levels and more stable than in HEPES buffered media, supporting the use of histidine buffered media for cooled transportation of human ovaries.

Future potential of in vitro maturation including fertility preservation

Summary: In several mammalian species, oocytes from small antral follicles after in vitro maturation (IVM) are successfully used for procreation. Humans are the exception, mainly due to limited access to immature oocytes and unique regulation of oocyte maturation. However, recent findings suggest that human IVM can be developed, with factors such as oocyte diameter, follicle size, hormone levels, and cumulus-oocyte complex secretion influencing the maturation process. Furthermore, studies on signal transduction pathways and hormone levels during physiological conditions provide new insights for improving the current IVM platform. With continued research efforts, it is envisioned that a human IVM platform equally effective as in other mammalian species can be achieved. (Fertil Sterile 2023;119:550-9. Summary: This study investigates the substances and signal transduction pathways involved in the regulation of human oocyte maturation downstream to the effects of FSH and LH. The results reveal that human oocyte maturation is a multifactorial process involving several different signal transduction pathways. Various pathways, including those involving CNP, EGF-related peptides, TGF-beta family members, activins/inhibins, FF-MAS, and MDK, are potentially active, while the GDF9 and BMP15 pathways show less pronounced influence.
