Summary: In this study, a novel heterozygous missense mutation in ZP2 was identified in a primary infertile female. The mutation was shown to reduce ZP2 expression and impede ZP2 secretion in cell lines. A mouse model with the mutation was constructed and it was found that the mutation caused degeneration of oocytes without zona pellucida (ZP) and resulted in complete infertility. ZP deletion also impaired folliculogenesis and caused female infertility in this mouse model. This study expands the spectrum of ZP2 mutation sites and increases the understanding of pathogenic mechanisms of ZP2 mutations.

Summary: Aneuploidy originating during meiosis in oocytes is a major cause of reduced fertility, implantation failure, and miscarriage in women beyond their mid-thirties. The underlying molecular defects leading to error-prone chromosome segregation in older women's oocytes are not well understood, but loss of chromosome cohesion, defective microtubule dynamics, and spindle assembly are major contributors. In aged mares, gene expression for the kinases Mps1, Spc25, and AurkC is reduced, leading to instability in spindle (re)assembly and predisposing to chromosome segregation errors.
