Summary: The study shows that LE is a feasible treatment option for early perianal ASCCs but not for anal canal ASCCs. Acceptable long-term outcomes can still be achieved with adjuvant therapy.

Summary: This study examined the genetic mutations in pretreatment tumor tissue and posttreatment ctDNA in patients with metastatic SCCA and assessed the impact of therapy on the concordance between them. The results showed a high degree of temporal mutational heterogeneity in patients with SCCA, supporting the use of ctDNA as a real-time tracking mechanism for solid tumors' molecular evolution. The study also highlighted the potential of ctDNA in identifying emerging actionable mutations. Further research is needed to validate these findings.

Summary: Anal canal adenocarcinoma is a rare malignant tumor of the intestinal tract with low clinical incidence. Diagnosis of this condition typically requires a combination of imaging techniques such as CT, MRI, colonoscopy, and histopathology. Comprehensive treatment including abdominal perineal resection, radiotherapy, and chemotherapy is crucial for managing anal canal adenocarcinoma.

Summary: The differences in molecular profiling between TP53-MT and TP53-WT SCCA suggest different carcinogenic pathways that may affect treatment response, while low-frequency mutations in several druggable genes could offer therapeutic opportunities for patients with SCCA.

Summary: This article reports a rare case of goblet cell adenocarcinoma in the anal canal with perianal Paget disease. The tumor in the anal canal was composed of signet-ring-like cells and produced copious mucin. Small groups of atypical neoplastic cells were present in the lesion of perianal Paget disease. The clinical diagnosis of this rare case is easily overlooked, highlighting the importance of recognizing this rare disease.

Summary: Esophageal basaloid squamous cell carcinoma (BSCC) exhibits a more clinically virulent course, and the expression of p53 and p63 are associated with prognosis in BSCC, suggesting that evaluation of multiple cancer biomarkers may be a promising auxiliary diagnostic indicator in BSCC.

Summary: Squamous cell carcinoma is a common malignant tumor in the anal canal, and FDG PET/CT is recommended for its nodal staging, radiotherapy planning, and response assessment. We present an interesting case of dual primary malignancy involving the anal canal and rectum, which was identified by F-18-FDG PET/CT and confirmed as synchronous squamous cell carcinoma through histopathology.

Summary: Stable cell lines derived from primary tissues and tumors are commonly used in medical research. This study evaluated gene expression profiles of esophageal tissues and cell lines, finding discrepancies in the gene expression profiles of Barrett's esophagus (BE) cell lines compared to primary BE tissues and esophageal adenocarcinoma (EAC) tumors. Exposure to an acidic bile environment did not alter the squamous-like gene expression pattern of a BE cell line. The findings underscore the importance of carefully selecting appropriate in vitro tools for studying esophageal adenocarcinoma.

Summary: Anal squamous cell carcinoma incidence is increasing, with 3.05% unexpectedly identified in hemorrhoidectomy specimens. Patients with hemorrhoidal squamous cell carcinoma had similar demographics and treatment outcomes compared to nonhemorrhoidal cases, but were more likely to have stage I disease. The study suggests a need for improved labeling and separate submission of hemorrhoid specimens to prevent potential overtreatment with chemoradiation.

Summary: The definition of the anal canal was revised in the TNM classification (8th edition). A multi-institutional study by the JSCCR in Japan revealed the characteristics of anal canal cancer (ACC), including the predominant types of carcinoma and the association with HPV infection. Prognosis analysis showed no significant difference in the 5-year overall survival rates between different treatment groups, but HPV-positive patients had better survival. Although an HPV vaccine for anal canal SCC has been approved internationally, it is urgently needed for men in Japan.

Summary: Our study on morular metaplasia (MM) in endometrial lesions shows that mixed MM/SD cases exhibit squamous differentiation (SD) mostly within MM areas, while conventional SD cases show a direct transition from glandular epithelium to SD. Immunohistochemistry results also demonstrate distinct expression patterns between the two types of cases. Further research is needed to define the nature of MM and its relationship with SD.
