Summary: A study found that luteolin (LUT), a natural component, can significantly protect the structure and function of liver grafts, prolong recipient rat survival, ameliorate T cell infiltration, and downregulate proinflammatory cytokines. LUT also inhibits CD4+ T cell proliferation and Th cell differentiation, while increasing the proportion of Tregs, which is crucial for its immunosuppressive effect. This discovery may have important implications for improving immunosuppressive regimens for organ transplantation.

We compared the immune response in heart/kidney transplantation rejection and liver transplantation tolerance. Summary: The study found that using 4-Methylumbelliferone (4MU) can alleviate acute allograft rejection in a murine model of lung transplantation, significantly improving transplant outcomes by reducing T cell-mediated lung inflammation, LMW-HA content, and pathology scores. Additionally, in vitro experiments showed that 4MU directly attenuated the activation, proliferation, and differentiation of naive CD4(+) T cells into Th1 cells. Given the safety of 4MU for human use, further research on its potential treatment for acute lung allograft rejection may have clinical significance.

Summary: In this study, we investigated the relationship between plasma levels of CXCL9/CXCL10 and histopathologic lung allograft injuries, as well as the risk of developing CLAD. We found that elevated plasma CXCL9/CXCL10 levels were associated with injury patterns associated with CLAD, acute rejection, and acute lung injury, and there was a dose-response relationship between chemokine levels and CLAD risk. Importantly, the presence of elevated plasma chemokines was necessary for the association between histopathologic injury and CLAD. Similar associations and interactions were observed with CXCL9/CXCL10 levels in bronchoalveolar lavage fluid. Elevated plasma CXCL9/CXCL10 during allograft injury may contribute to CLAD development and could be a potential minimally invasive biomarker for immune monitoring.

Summary: Chronic lung allograft dysfunction (CLAD), characterized by decline in pulmonary function and bronchiolitis obliterans, lacks approved therapies. Single-cell RNA-Seq and spatial transcriptomic analysis identified the important role of JAK-STAT signaling in airway epithelial cells in CLAD. Activation of JAK-STAT signaling leads to upregulation of MHC-I in airway basal cells, resulting in cytotoxic T cell-mediated basal cell death. This study provides mechanistic insight and potential therapeutic strategy for CLAD.

Summary: Retinal pigment epithelium (RPE) cell allotransplantation is a potential solution for retinal diseases. However, the RPE-complement system triggered by CL-11 could hinder the success of transplantation due to the inflammatory response and T cell recognition promotion. We found that CL-11 inhibited T cell proliferation by binding to T cells and downregulating CD28 expression. Additionally, soluble CL-11 secreted by stressed RPE cells may contribute to the immunosuppressive properties of RPE cells.

Summary: This study suggests that the donor-derived cell-free DNA percentage at the time of respiratory viral infection could be a predictor of chronic lung allograft dysfunction progression in lung transplant recipients. The recipients with higher levels of %ddcfDNA experienced greater decline in lung function and had a higher risk of progression and failure within one year.
