A Role for Resistin in the Ovary During the Estrous Cycle

In a previous study, we showed that resistin expression increased during ovarian follicle development in prepubertal pigs and had direct effects on steroidogenesis, suggesting an important role for resistin in the ovary during puberty. To determine its potential regulatory role in the ovary during the estrous cycle, using real-time PCR, immunoblotting, immunohistochemistry, and ELISA methods, we quantified the expression, immunolocalization and concentration of resistin in different sized ovarian follicles (small, 2-4 mm; medium, 4-6 mm; and large, 8-12 mm) in mature pigs. We then determined the effects of recombinant resistin (0.1, 1, and 10 ng/ml) on steroid hormone (progesterone-P4, androstendione-A4, testosterone-T, and estradiol-E2) secretion and steroidogenic enzyme (3 beta HSD, CYP17A1, 17 beta HSD, and CYP19A1) gene and protein expression in ovarian follicles. We found no differences in the resistin expression between all of the examined follicles. Immunostaining analysis also showed resistin expression in the cytoplasm of both granulosa and theca cells, where it was localized more abundantly in the granulosa cells compared to the theca cells. Recombinant resistin direct stimulated P4, A4, and T secretion via increased expression of 3 beta HSD, CYP17A1, and 17 beta HSD, suggesting an autocrine and/or paracrine regulatory role in the porcine ovary during the estrous cycle.

Summary: This study aimed to determine the concentrations of polycyclic aromatic hydrocarbons (PAHs), especially naphthalene, in the plasma and adipose tissue of French and Polish bariatric patients and investigate their correlation with body mass index (BMI), adipokines expression levels, and the effects of naphthalene on cell proliferation and differentiation. The results showed that PAHs and naphthalene could potentially be obesogenic molecules, as naphthalene concentration in adipose tissue was positively correlated with BMI and adipokines expression in French patients. Moreover, naphthalene exposure increased cell proliferation and differentiation, suggesting an involvement in adipogenesis/lipogenesis.

Expression and regulation of visfatin/NAMPT in the porcine corpus luteum during the estrous cycle and early pregnancy

Spexin role in human granulosa cells physiology and PCOS: expression and negative impact on steroidogenesis and proliferation

Summary: This study investigates the expression and regulatory function of the novel neuropeptide Spexin (SPX) in the human ovary, and its potential role in polycystic ovarian syndrome (PCOS) pathogenesis. The results show that SPX levels are decreased in obese and PCOS women, and SPX inhibits granulosa cell (GC) proliferation and steroidogenesis. GALR2/3 and kinases MAP3/1, AKT, and STAT3 are involved in mediating these effects.

Phoenixin-14 as a novel direct regulator of porcine luteal cell functions

Summary: This study investigated the expression of phoenixin-14 in porcine corpus luteum and its impact on endocrine function. The results showed that phoenixin-14 can regulate the secretion of progesterone and estradiol and affect the expression of hormone receptors in luteal cells. Moreover, G protein-coupled receptor 173 and extracellular signal-regulated kinase 1/2 play important roles in the regulation of endocrine function in luteal cells.

Summary: Recent studies have shown that visfatin is involved in the regulation of female reproduction. This study aimed to investigate the expression and localization of visfatin in the ovarian follicles of prepubertal and mature gilts, and the effects of various hormones on visfatin levels. The results showed that visfatin expression is dependent on the maturity of the animals and the stage of ovarian follicle development. The study also revealed that visfatin is regulated by different hormones and may play a crucial role in porcine ovarian follicular function.

Summary: Visfatin is expressed in both the anterior and posterior lobes of the pig pituitary, and its expression is influenced by the estrous cycle and early pregnancy. GnRH, FSH, LH, and insulin can stimulate the secretion of Visfatin in pituitary cells, with LH specifically enhancing Visfatin release during days 2 to 3 of the cycle.

Summary: This study aimed to investigate the impact of VD3 supplementation on RARRES2 and ADIPOQ levels in the uterus of PCOS rats. The findings showed that VD3 supplementation restored the levels of these adipokines and increased the expression of their receptors in the uterus of PCOS rats, indicating a new mechanism of VD3 action in uterine physiology.

Synergistic Action of MCL-1 Inhibitor with BCL-2/BCL-XL or MAPK Pathway Inhibitors Enhances Acute Myeloid Leukemia Cell Apoptosis and Differentiation

Summary: Acute myeloid leukemia (AML) is a blood cancer characterized by abnormal cell proliferation and inhibition of apoptosis. The MCL-1 protein plays a critical role in the survival of AML cells. This study investigated the effects of a specific MCL-1 inhibitor, S63845, alone and in combination with other inhibitors, on AML cell lines. The results showed that S63845 decreased cell viability and induced apoptosis, and combined treatment enhanced apoptosis and induced cell differentiation. These findings suggest the potential use of MCL-1 inhibitors in combination with other pro-survival protein inhibitors.

The results showed fluctuation in the expression of OMNT1 in different stages of the cycle and differences in expression levels between pig breeds. Effect of immunocastration using Improvac on the regulation of adiponectin and leptin in the testes of Landrace boars

Summary: This study revealed, for the first time, the effects of GnRH vaccination on the testicular function of Landrace pigs. It showed disrupted adiponectin and leptin signaling, increased cholesterol concentration, and attenuated spermatogenesis due to halted androstenone production. The altered GnRH signaling may impact the adipokine system in the testes, leading to further functional changes and lipid metabolism disturbances.

Summary: The prevalence of adult obesity has increased significantly, and the endocrine system plays a crucial role in energy balance and fat deposition. Endocrine disruptor chemicals (EDCs) have been extensively studied for their association with obesity. EDCs can trigger obesity through various mechanisms, such as influencing adipocyte differentiation, increasing triglyceride storage, or elevating adipocyte number. Furthermore, EDCs and obesity may affect reproductive functions and lead to conditions like polycystic ovarian syndrome and decreased sperm motility.
