An efficient one-pot, three-component synthesis method of highly functionalized coumarin fused 7,13-dihydro-5-oxa-13-aza-indeno[2,1-b]phenanthrene and chromeno[4,3-b]quinoline derivatives was accomplished using aromatic aldehyde, 4-aminocoumarin and cyclic 1,3-dione in acetic acid at reflux temperature. Environmental-friendly protocol has advantages of high yields, creating three new bonds and one stereocenter in single operation. Further, XRD and DFT study confirmed the molecular structure, stability and different interaction in the crystal packing.

On the inhibition of cytochrome P450 3A4 by structurally diversified flavonoids

Summary: The study revealed that chrysin has the largest inhibitory effect on CYP3A4, while quercetin and flavopiridol serve as representative examples of structurally different flavonoids. The inhibition parameters for these compounds were evaluated using the calibrated QM/MM strategy, aiding in quantitatively understanding the functional behavior of the whole flavonoid family. Additionally, a kinetic threshold for further assessment of the time-dependent inhibition of CYP3A4 by flavonoids was explored.

On the recognition of Yersinia protein tyrosine phosphatase by carboxylic acid derivatives

Summary: This study provides insights into the structural characteristics of YopH protein in pathogenic conditions and offers a basis for designing effective YopH inhibitors. By using molecular docking simulations and a quantum mechanics/molecular mechanics model, the conformational changes induced by aurintricarboxylic acid are investigated. The active/inactive functional behavior of YopH is explored, and the implications for experimental research are discussed.
