Summary: This article describes the pharmacological activities of two highly potent and selective ATM inhibitors, M3541 and M4076. The inhibitors were found to suppress DSB repair, clonogenic cancer cell growth, and enhance the antitumor activity of radiotherapy in cancer cell lines. In vivo experiments showed that oral administration of these inhibitors in mice with human tumor xenografts led to complete tumor regressions. Additionally, they demonstrated strong combination potential with PARP and topoisomerase I inhibitors.

Summary: PARP inhibitors have been shown to effectively treat natural killer/T cell lymphoma (NKTCL) by inhibiting cell proliferation, promoting apoptosis, and inducing cell cycle arrest. The combination of PARP inhibitors and cisplatin demonstrated synergistic effects both in vitro and in vivo. LMO2 deficiency reduced the sensitivity of NKTCL cells to PARP inhibitors.

Summary: This review discusses the current and investigative strategies for targeting DNA repair in the management of glioma, highlighting the importance of generating DNA damage and inhibiting repair mechanisms to achieve cytotoxicity. Glioma biomarkers such as IDH mutation and MGMT promoter methylation may provide specific susceptibility to DNA damage and repair inhibition, and targeting DNA repair mechanisms with novel agents shows promise as an approach to glioma treatment. Ongoing investigations are focused on optimizing this approach in the clinical setting.

Summary: AMOT is a membrane protein aberrantly expressed in solid tumors, and its reduced expression in DLBCL is associated with poor clinical prognosis. Overexpression of AMOT in DLBCL cells inhibits cell viability, increases sensitivity to doxorubicin, and reduces activation of key kinases for the DNA damage response, suggesting it acts as a tumor suppressor via inhibition of DDR. AMOT may be a novel potential target for therapeutic intervention in DLBCL.

Summary: This study aimed to evaluate the safety and recommended dose of olaparib in combination with alpelisib in patients with breast cancer, and explore its effects on different subtypes. The results showed that this combination therapy was tolerable for pre-treated TNBC patients and demonstrated activity in non-BRCA mutant patients. Analysis of circulating-free DNA also provided important prognostic information.

Summary: Although major progress has been made in mantle cell lymphoma (MCL) therapeutics, MCL still has a poor prognosis with a median survival of less than four years. Several genes, including NOTCH1 and NOTCH2, have been found to be mutated in MCL, leading to upregulation of genes involved in angiogenesis, cell cycle progression, and cell migration and adhesion. Mutated NOTCH genes are associated with aggressive features in MCL and inferior survival. This article discusses the role of NOTCH signaling in MCL biology and ongoing efforts for targeted therapeutic interventions.
