Poster summary
- Intranasal administration offers an alternative way for brain targeting. In this project, we studied the design of two innovative mucoadhesive nanoformulations for BNN27 intranasal delivery. Negatively-charged and uncharged liposomes were coated with chitosan (CHT) at different CHT/lipid ratios. Medium MW CHT-coated PC/PG/BNN27 liposomes at 0.1w/w CHT/lipid ratio demonstrated highest coating efficiency & mucoadhesive properties. Furthermore, preparation of mucoadhesive nanoemulsions (MNEs) was studied. NEs were prepared by the spontaneous emulsification method using Capmul MCM as oily phase. MNEs with 10%w/w Capmul MCM and 0.3%w/w CHT, displayed optimal physicochemical characteristics. Optimal nanoformulations were evaluated for their permeability across a cellular BBB model (hCMEC/D3 cell monolayers) in vitro. Also, their in vivo fate after intranasal administration revealed the superiority of CHT-NEs for more efficient BNN27 distribution to the brain, compared to CHT-LIPs. Current results prove that CHT-NEs succeed to deliver more BNN27 to the brain (and faster), compared to CHT-coated LIPs.
