Summary: PTEN is a negative regulator of the INS-PI3K-AKT pathway and plays a critical role in metabolism and cell growth. It is frequently mutated in cancer, but overexpression of PTEN extends lifespan in mice. Our recent study demonstrated that PTEN activates chaperone-mediated autophagy (CMA) and selectively degrades metabolic enzymes to suppress glycolysis and fatty acid synthesis. This suggests that PTEN-mediated CMA activation could modulate health and metabolism.

Summary: The study reveals the crucial role of COPII vesicle-related protein TFG in autophagy induction, affecting ULK1 puncta number and localization, isolation membrane formation, and correct dynamics of LC3B and ATG proteins association for proper omegasome and autophagosome formation. The interaction between TFG and ATG8 protein LC3C through a canonical LIR motif is essential for TFG's activity in autophagy, favoring LC3C-ULK1 binding. Overall, TFG is identified as a molecular scaffold linking the early secretion pathway to autophagy.

FXR in the liver and intestine plays a crucial role in regulating postprandial nutrient disposal.
