Summary: T cells are crucial for generating effective responses against microbial pathogens and tumor surveillance, but unwanted T cell activation can lead to harmful inflammation. Tight regulation by transcription factors, epigenetic regulators, and miRNAs is necessary for optimal T cell responses. Recent progress has identified key immune regulatory miRNAs controlling T cell subset differentiation and function, as well as the extrinsic role of miRNAs in regulating T cell biology through interactions with immune and non-immune cell populations. Important questions regarding the complex nature of miRNA-mediated control of T cell immunity still need further investigation.

Summary: Gamma delta T cells, a unique subgroup of T cells, can promote anti-tumor immune response but also potentially contribute to tumor progression. Studies have summarized and discussed their functions and effects in cancer, as well as reviewed recent anti-tumor immunotherapy based on gamma delta T cells, and identified existing problems and future prospects of this immunotherapy.

Summary: In a septic mouse model, aggravated pyroptosis was observed in CD4+ T cells, accompanied by impaired autophagy activity and an overactivated mTOR signaling pathway. Depletion of mTOR relieved autophagy deficiency and reduced the proportion of pyroptotic CD4+ T cells. This indicates that autophagy negatively regulates pyroptosis in CD4+ T cells, which is under the control of the mTOR pathway.

NHR-49 is differentially regulated by interventions that bring about long-term changes (life span extension) versus short-term stress (pathogen exposure) and orchestrates discrete outputs, including pathogen-specific transcriptional programs.

Summary: Activation of the IRE1/XBP1s axis in cancer promotes immunosuppression and tumor growth. This study investigated the role of IRE1/XBP1s in dendritic cells (DCs) in tumors, specifically in conventional type 1 DCs (cDC1s), which are important targets in immunotherapy. The results show that deletion of the RNase domain of IRE1 and/or XBP1s in DCs and cDC1s does not affect tumor growth or T cell responses. These findings argue against a pro-tumorigenic role of the IRE1/XBP1s pathway in tumor-associated DC subsets.

Summary: Activation of STING signaling enhances the differentiation and antitumor functions of T(H)1 and T(H)9 cells by increasing their production of IFN-gamma and interleukin-9, respectively. IRF3 and IFNARs are required for the enhancement of T(H)1 cell differentiation, while mTOR signaling is implicated in T(H)9 cell differentiation in response to STING activation. STING activation boosts the antitumor activity of T(H)1 and T(H)9 cells through their cytokine production, and enhances the efficacy of T(H)9 cells in adoptive transfer against melanoma.

Summary: A hostile microenvironment in tumor tissues disrupts endoplasmic reticulum homeostasis and activates the unfolded protein response. The UPR component X-box binding protein 1 (XBP1) in cancer cells promotes the synthesis and secretion of cholesterol, which activates myeloid-derived suppressor cells (MDSCs) and causes immunosuppression. Inhibition of XBP1 or reduction of tumor cholesterol content significantly reduces MDSC abundance and triggers robust anti-tumor responses.

Summary: This study proposed a strategy to enhance SARS-CoV-2 specific T cell functionality by fusing autophagosome-associated LC3b protein to the nucleocapsid (N). The N-LC3b protein showed improved targeting and elicited stronger CD4(+) and CD8(+) T cell immune responses, including higher frequency of polyfunctional T cells and improved T cell proliferation. Additionally, the strategy also induced a robust humoral immune response. These findings provide insights for the development of a promising universal vaccine against SARS-CoV-2 variants and other emerging infectious diseases.
