Prolyl endopeptidase (E.C. 3.4.21.26, PREP) also known as prolyl oligopeptidase is an enzyme which cleaves several peptides at the carboxyl side of proline residues. Since brain contains relatively large amounts of this enzyme and because of its substrate specificity it has been suggested to play a role in the metabolism of neuropeptides, acting both on their maturation and their degradation. The final step of neuropeptide processing occurs in the synaptic vesicles and degradation of most of these peptides takes place in the synaptic cleft. Thus, a localization of PREP in these cellular compartments appears to be feasible. Here we summarize recent data and provide novel evidence for the subcellular localization of PREP. Most importantly, immunocytochemical double labelling, confocal laser scanning and electron microscopic procedures as well as functional assays strongly suggest a role for PREP in intracellular transport mechanisms and in protein secretion.

Aggrecan modulates the expression and phosphorylation of tau in a novel bigenic TauP301L-Acan mouse model

Sophie Schmidt, Caroline Stapf, Sandra Schmutzler, Ingolf Lachmann, Thomas Arendt, Max Holzer, Mandy Sonntag, Markus Morawski

Summary: Selected neurons in the central nervous system are surrounded by perineuronal nets (PNs) which regulate plasticity and synaptic physiology. Research showed that aggrecan, a component of PNs, has a neuroprotective role by preventing internalization of pathological tau forms. However, in a mouse model, reduced aggrecan levels led to increased tau protein and phosphorylation, suggesting a correlation between aggrecan and tau pathology triggered by mutation.

miR24-3p activity after delivery into pancreatic carcinoma cell lines exerts profound tumor-inhibitory effects through distinct pathways of apoptosis and autophagy induction

Summary: The replacement of miR24-3p may be an effective strategy for pancreatic cancer therapy, leading to cell inhibition through the induction of apoptosis, autophagy, and ROS. Comprehensive analyses revealed the transcriptional regulation of several important oncogenes and oncogenic pathways by miR24-3p on the molecular level.

High Iron and Iron Household Protein Contents in Perineuronal Net-Ensheathed Neurons Ensure Energy Metabolism with Safe Iron Handling

Anja Reinert, Tilo Reinert, Thomas Arendt, Markus Morawski

Summary: A subpopulation of neurons, characterized by a special extracellular matrix composition forming a perineuronal net (PN) with a high affinity to iron, is less vulnerable to iron-induced oxidative stress and neurodegeneration. These PN+ neurons have higher intracellular iron concentrations and increased levels of iron proteins, with the iron concentration of the PN itself not being increased. The properties of these resilient pacemaker neurons with bustling iron metabolism and outstanding iron handling capabilities contribute to their low vulnerability to iron-induced oxidative stress and degeneration.

Finding the best clearing approach- Towards 3D wide-scale multimodal imaging of aged human brain tissue

Henriette Rusch, Malte Brammerloh, Jens Stieler, Mandy Sonntag, Siawoosh Mohammadi, Nikolaus Weiskopf, Thomas Arendt, Evgeniya Kirilina, Markus Morawski

Summary: The accessibility of new wide-scale multimodal imaging techniques has led to the emergence of numerous tissue clearing techniques. This study compares seven different tissue clearing approaches for aged human brain tissue blocks (>5 mm) to identify the most practical and efficient method. The results show that combining the CLARITY or iDISCO techniques with specific imaging setups is the best approach for clearing aged human brain tissue and generating 3D microscopic images.

Adhesion GPCR GPR56 Expression Profiling in Human Tissues

Fyn Kaiser, Markus Morawski, Knut Krohn, Nada Rayes, Cheng-Chih Hsiao, Marianne Quaas, Gabriela Aust

Summary: GPR56, also known as gene ADGRG1, plays significant roles in various physiological processes and shows broad expression in human tissues, especially in microglia in the adult brain and cuboidal secreting epithelia in peripheral tissues. High levels of ADGRG1 mRNA in specific tissues correlate with elevated levels of GPR56 protein expression in corresponding cell types.

Towards a representative reference for MRI-based human axon radius assessment using light microscopy

Summary: Non-invasive assessment of axon radii via MRI is important for clinical and neuroscience research, but lacks representative histological reference data. This study proposes a method using CNN-based segmentation to generate a representative reference for the effective radius (r(eff)) of axons. The results show that r(eff) estimation is more accurate and less biased compared to the traditional method of estimating the arithmetic mean radius (r(arith)).

Tau Protein Modulates Perineuronal Extracellular Matrix Expression in the TauP301L-acan Mouse Model

Sophie Schmidt, Max Holzer, Thomas Arendt, Mandy Sonntag, Markus Morawski

Summary: Tau mutations promote the formation of tau oligomers and filaments, while certain types of neurons are spared of tau pathology due to the protective function of perineuronal nets (PNs), which prevent the internalization of misfolded tau. The study demonstrates a complex interrelation between tau and the proteoglycans of the PN, suggesting that tau can regulate the structure and protein expression of PNs.

High-resolution magnetization-transfer imaging of post-mortem marmoset brain: Comparisons with relaxometry and histology

Henrik Marschner, Andre Pampel, Roland Mueller, Katja Reimann, Nicolas Bock, Markus Morawski, Stefan Geyer, Harald E. Moeller

Summary: Cell membranes and macromolecules/paramagnetic compounds interact with water proton spins, providing MRI contrast and tissue composition information. Quantitative magnetization transfer (qMT) parameters were measured at high spatial resolution in a fixed marmoset brain slice and compared to histology results. R1obs and R2* were linearly correlated with iron content, while distinct differences were observed between gray and white matter for qMT parameters and relaxometry. These findings suggest that the water interacting macromolecular pool consists of myelin and non-myelin contributions, and R1obs and R2* are more sensitive for depicting microstructural differences between cortical layers than T.
