Recruitment of host's progenitor cells to sites of human amniotic fluid stem cells implantation

The amniotic fluid is a new source of multipotent stem cells with a therapeutic potential for human diseases. Cultured at low cell density, human amniotic fluid stem cells (hAFSCs) were still able to generate colony-forming unit-fibroblast (CFU-F) after 60 doublings, thus confirming their stamina] nature. Moreover, after extensive in vitro cell expansion hAFSCs maintained a stable karyotype. The expression of genes, such as SSEA-4, SOX2 and OCT3/4 was confirmed at early and later culture stage. Also, hAFSCs showed bright expression of mesenchymal lineage markers and immunoregulatory properties. hAFSCs, seeded onto hydroxyapatite scaffolds and subcutaneously implanted in nude mice, played a pivotal role in mounting a response resulting in the recruitment of host's progenitor cells forming tissues of mesodermal origin such as fat, muscle, fibrous tissue and immature bone. implanted hAFSCs migrated from the scaffold to the skin overlying implant site but not to other organs. Given their in vivo: (i) recruitment of host progenitor cells, (ii) homing towards injured sites and (iii) multipotentiality in tissue repair, hAFSCs are a very appealing reserve of stem cells potentially useful for clinical application in regenerative medicine. Summary: This study generated a novel ADC called Cet-ZA, which can specifically target CRC. Importantly, patient-derived CRC organoids or CRC tumor cell suspensions could induce the expansion of Vδ2 T cells from peripheral blood and tumor infiltrating lymphocytes when primed with Cet-ZA, and Cet-ZA can promote Vδ2 T cell-mediated killing of CRC organoids.

Summary: The advent of immune checkpoint inhibitors (ICIs) has dramatically changed the treatment of non-small cell lung cancer (NSCLC). However, their efficacy remains limited to a subset of patients. This study evaluated the prognostic role of soluble (s) immune checkpoints and immune checkpoints associated with extracellular vesicles (EVs) in NSCLC patients treated with ICIs. The results showed that high levels of sPD-L1, sB7-H3, and sB7-H4 were associated with different prognoses in advanced NSCLC patients treated with ICIs. The study also suggested potential involvement of EVs in the disease response to ICIs.

Pilot study investigating BP-180 in extracellular vesicles derived from blister fluid of bullous pemphigoid patients

Summary: This study aimed to investigate the presence of BP targeted autoantigens in blister fluid-derived extracellular vesicles (EVs). EVs were isolated from blister fluid of BP patients and suction blister fluid of healthy donors, and EV characterization was performed. Western blot analysis revealed the presence of BP180 in EVs derived from blister fluid of 3 out of 6 BP patients, while no BP180 was detected in EVs from healthy donors. This discovery provides insights into the pathogenesis of BP.
