High-Resolution Amplified Fragment Length Polymorphism Typing of Lactococcus lactis Strains Enables Identification of Genetic Markers for Subspecies-Related Phenotypes

A high-resolution amplified fragment length polymorphism (AFLP) methodology was developed to achieve the delineation of closely related Lactococcus lactis strains. The differentiation depth of 24 enzyme-primer-nucleotide combinations was experimentally evaluated to maximize the number of polymorphisms. The resolution depth was confirmed by performing diversity analysis on 82 L. lactis strains, including both closely and distantly related strains with dairy and nondairy origins. Strains clustered into two main genomic lineages of L. lactis subsp. lactis and L. lactis subsp. cremoris type-strain-like genotypes and a third novel genomic lineage rooted from the L. lactis subsp. lactis genomic lineage. Cluster differentiation was highly correlated with small-subunit rRNA homology and multilocus sequence analysis (MLSA) studies. Additionally, the selected enzyme-primer combination generated L. lactis subsp. cremoris phenotype-specific fragments irrespective of the genotype. These phenotype-specific markers allowed the differentiation of L. lactis subsp. lactis phenotype from L. lactis subsp. cremoris phenotype strains within the same L. lactis subsp. cremoris type-strain-like genomic lineage, illustrating the potential of AFLP for the generation of phenotype-linked genetic markers.

The lack of a gold standard for Ng infection confirmation presents a dilemma in public health settings: choosing between missed diagnoses or overtreatment.

Assessing the Impact of Diet on the Mucosa-Adhered Microbiome in Piglets Using Comparative Analysis of Rectal Swabs and Colon Content

Summary: This study investigated the impact of diet on the mucosa-adhered microbiota in pre-weaning piglets by analyzing rectal swabs and colon samples. The results showed variable enrichment of adhered populations in rectal swabs and suggested that early life feeding may suppress post-weaning diarrhea. Furthermore, the paired swab-colon microbiota information can be used to predict the adhered microbiota in rectal swab samples.

Summary: This study investigated the travel history and behavior of patients and their correlation to highly resistant microorganism (HRMO) carriage upon hospital admission. It found that half of the admitted patients had traveled abroad in the last year, with only a small percentage traveling outside Europe. The majority of patients were aware of the risk of HRMO carriage during international travel and supported screening strategies to minimize the risk of introducing HRMO.

Summary: This study demonstrates the importance of detailed whole genome sequencing analysis for highly refined epidemiological analysis of carbapenem non-susceptible Pseudomonas aeruginosa. The analysis revealed variations in transmission and acquisition of CNPA strains among patients, as well as the correlation between genomic sequence changes and the distribution of antibiotic resistance and virulence genes.

Contact tracing for vancomycin-resistant Enterococcus faecium (VRE): evaluation of the Dutch policy of quintuple screening cultures

Summary: This study aimed to improve the sensitivity of detecting vancomycin-resistant Enterococcus faecium (VRE) transmission and determine the time from presumed exposure to detectable colonization. Retrospective analysis of 9 VRE outbreaks showed that using the first three out of five rectal swabs identified 89% of all secondary cases, compared to only 64% with the first swab. In addition, taking one or more rectal swabs around day 9 after presumed exposure was found to be the most effective screening approach.

Environmental contamination with highly resistant microorganisms after relocating to a new hospital building with 100% single-occupancy rooms: A prospective observational before-and-after study with a three-year follow-up

Summary: This study aimed to compare the environmental contamination with highly resistant microorganisms (HRMO) between an old hospital building and a new hospital building, and to investigate the environmental contamination in the new building after relocation. The results showed a significant decrease in HRMO contamination in the new building, which is important for hospital design and construction considerations.
