Quantitative trait loci (QTL) affecting growth traits have previously been mapped in linkage groups (LG) 2, 3 and 23 of Barramundi (Lates carcalifer), but these QTL have not been verified in different genetic backgrounds and environments. Here, we report the identification and verification of QTL for growth traits on LG2, 3, 10 and 23 in F-1 families constructed using brooders from the Singapore Marine Aquaculture Center (MAC) and from wild stocks collected in Thailand (THAI). The previously detected QTL for body weight and length linked to marker Lca371 on LG2 were confirmed in both the MAC and THAI families, whereas other QTL previously mapped to LG3 and 23 were only detected in one of the two families. QTL for body weight and length were identified in the MAC family, but not in the THAI family, in a region where the insulin-like growth factor 2 (IGF2) and tyrosine hydroxylase 1 (TH1) genes are located on LG10. Significant epistatic interactions were identified between markers Lca287 on LG2 and IGF2 on LG10 for growth trait QTL in the MAC family, but not in the THAI family. Effects of the IGF2, TH1 and parvalbumin 1 candidate genes were family-specific. Our results indicate that some but not all QTL are family-specific in Barramundi.

EgSPEECHLESS Responses to Salt Stress by Regulating Stomatal Development in Oil Palm

Zhuojun Song, Le Wang, Chongcheong Lai, May Lee, Zituo Yang, Genhua Yue

Summary: In this study, the physiological responses and underlying mechanisms of oil palm to salt stress were investigated using RNA-Seq analysis. The results showed that salt stress activated the expression of EgSPCH gene and increased stomatal density in oil palm. Additionally, several differentially expressed genes were found to be involved in salt response-related bioprocesses. Overexpression of EgSPCH in Arabidopsis resulted in increased stomatal production and decreased salt tolerance.

The Insertion in the 3′ UTR of Pmel17 Is the Causal Variant for Golden Skin Color in Tilapia

Feng Liu, Fei Sun, Gang Qiao Kuang, Le Wang, Gen Hua Yue

Summary: Understanding the relationship between genotypes and phenotypes is a key problem in biology. This study demonstrates that the Pmel17 gene is responsible for determining blackish and golden colors in fish, and identifies an insertion in the 3'UTR of Pmel17 as the causative mutation for golden coloration.

Occurrence of dorsal fin and opercular deformities and their effects on body weight in Asian seabass

Gen Hua Yue, Yan Fei Wen, Fei Sun, Le Wang, Zituo Yang, Hong Yan Pang

Summary: The study found that dorsal fin and opercular deformities are common in Asian seabass hatcheries. These anomalies not only affect appearance but also reduce growth performance. Molecular parentage analysis indicates that the heritabilities of these deformities might be low.

Characterization of pIgR and its association with resistance to iridovirus in Asian seabass

Yi Xuan Tay, Yepin Yu, Zituo Yang, Le Wang, Fei Sun, Gen Hua Yue

Summary: Understanding the molecular mechanisms of disease resistance is crucial for improving aquaculture. The pIgR gene was found to play an important role in the mucosal immunity of Asian seabass, inhibiting iridovirus infection. Two SNPs in the pIgR gene may be associated with resistance to iridovirus.

The evolution and expression of stomatal regulators in C3 and C4 crops: Implications on the divergent drought tolerance

Zhuojun Song, Le Wang, May Lee, Gen Hua Yue

Summary: Drought stress is a significant environmental hazard, and plants respond to it differently. C3 and C4 plants exhibit different cellular responses to drought, which may be associated with the divergent expression of their SPEECHLESS genes. Our research investigated the drought tolerance of C3 and C4 plants through gene expression analysis and stomatal assay under drought stress.

Combination of GWAS and FST-based approaches identified loci associated with economic traits in sugarcane

Le Wang, Shadame Yeo, May Lee, S. Endah, N. A. Alhuda, G. H. Yue

Summary: This study aimed to identify DNA markers associated with sugar content, stalk diameter, and sugarcane top borer resistance. The sugarcane samples were genotyped using RADseq technology. Through F-ST analysis and GWAS, a total of 9, 23, and 9 DNA variants were identified for sugar content, stalk diameter, and sugarcane top borer resistance, respectively. These markers have the potential to accelerate genetic improvement in sugarcane breeding.

Status of conventional and molecular breeding of salinity-tolerant tilapia

G. H. Yue, K. Y. Ma, J. H. Xia

Summary: Aquaculture is crucial for global food security. Developing species and varieties of omnivorous fish that can tolerate high salinity will enhance fish production. Certain tilapia species, such as Oreochromis mossambicus and Oreochromis aureus, are highly tolerant to salinity and have been used in aquaculture production in brackish and seawater. Different breeding methods, both conventional and molecular, have been implemented to improve the growth rate and salinity tolerance of tilapia. Advances in genomic resources and disruptive technologies provide optimism for novel breeding approaches to further enhance the production of salinity-tolerant tilapia.
