Genetic and environmental factors underlying variation in yield performance and bioactive compound content of hot pepper varieties (Capsicum annuum) cultivated in two contrasting Italian locations

Hot pepper (Capsicum spp.) is an economically considerable crop, particularly appreciated for its nutritional properties and antioxidants content. Levels of the latter depend on several factors including the cultivar and the environment. In the present study, diversity in morpho-agronomic traits and phytochemical compounds was investigated in 14 hot pepper accessions cultivated in two different pedoclimatic locations. The main source of variation was due to the genotype which showed significant differences for all traits at both locations. With regard to the bioactive compounds, the environment accounted for over 30% of variation for carotenoids, ascorbic acid and tocopherols and less than 0.5% for capsaicinoids. A strong genotype x environment interaction (p < 0.01) was observed in all traits with the exception of those related to fruit weight and shape. Genotyping by sequencing was used for molecular analysis revealing a total of 2120 single-nucleotide polymorphisms. Diversity on the basis of hierarchical clustering and structure analysis elucidated the relationships among accessions on the basis of their biological status and common ancestors, suggesting their contribution to the phenotypic performances observed. The comprehensive approach of this study provides useful information on the variability of phytochemicals mediated by the environment, revealing the challenge related to the genetic improvement of bioactive compounds. The integration of phenotypic and molecular data provides information to identify interesting hot pepper accessions to select for breeding programmes.

Resistance against broomrapes (Orobanche and Phelipanche spp.) in vegetables: a comprehensive view on classical and innovative breeding efforts

Summary: Broomrapes are root parasites that obtain nourishment from host plants, causing severe damage to vegetable crops worldwide. Traditional agronomic practices have proven to be ineffective in managing broomrapes. This review provides an update on the genetic and molecular mechanisms underlying host-parasite interactions, as well as an overview of breeding efforts for resistance to broomrapes in vegetables. Natural sources of resistance have been discovered through germplasm exploration, and artificial mutagenesis has provided additional variability. Advances in parasitic and host plant genomics, along with new breeding technologies, offer promising opportunities for future developments. Integration of different genetic resistance mechanisms, combined with innovative agronomic management practices, may result in a more effective and durable containment strategy.

Aquaphotomic, E-Nose and Electrolyte Leakage to Monitor Quality Changes during the Storage of Ready-to-Eat Rocket

Summary: This study evaluated the maintenance of freshness in ready-to-eat rocket salad packed under modified atmospheres using methods such as E-nose, electrolyte leakage test, NIR spectroscopy, and Aquaphotomics. The results showed that a gas mixture of 30% O-2 and 70% N-2 was the most effective in maintaining product freshness. NIR spectroscopy successfully grouped the samples based on storage time, while Aquaphotomics was able to detect changes in water structure during storage.

Variation of Bio-Morphometric Traits and Antioxidant Compounds of Brassica oleracea L. Accessions in Relation to Drought Stress

Genetic Analyses of Seed Longevity in Capsicum annuum L. in Cold Storage Conditions

Summary: Seed longevity is an important trait in genebank management, and little is known about the genetic basis of this trait in Capsicum. In this study, 1152 Capsicum accessions stored for 40 years were evaluated for seed longevity using SNP markers. A total of 224 marker trait associations were identified, and several candidate genes were discussed.

Development and application of Single Primer Enrichment Technology (SPET) SNP assay for population genomics analysis and candidate gene discovery in lettuce

Summary: Single primer enrichment technology (SPET) is a high-throughput genotyping method based on short-read sequencing of specific genomic regions harboring polymorphisms. It allows for efficient and reproducible genotyping of target loci and the discovery of closely linked, novel SNPs. In this study, the effectiveness of SPET was demonstrated through the design and application of a panel in lettuce, which identified genetic subgroups and revealed the differentiation of lettuce types from different regions. The potential of SPET for gene discovery was also demonstrated through genome-wide association analysis of agricultural traits in lettuce.

Application of High-Resolution Melting and DNA Barcoding for Discrimination and Taxonomy Definition of Rocket Salad (Diplotaxis spp.) Species

Summary: In this study, the genetic diversity of 17 Diplotaxis species was investigated using 5 DNA barcode markers, which revealed their taxonomic relationships. The combination of real-time PCR and high-resolution melting analysis proved effective in resolving taxonomic relationships and species identification. The findings demonstrate the effectiveness of this approach for taxonomic and genetic diversity studies.
