Summary: This study identified and analyzed the functional role of ATG4C, a protein involved in autophagy, in grass carp. ATG4C was found to be widely distributed in grass carp tissues, with high expression in the intestine. It showed a strong response in immune tissues after infection with grass carp reovirus (GCRV), indicating its involvement in the immune response. Additionally, the study confirmed the induction of autophagy and apoptosis by GCRV invasion, and revealed that ATG4C positively regulated autophagy while restricting apoptosis, leading to inhibition of GCRV replication.

Summary: In this study, it was found that the TBK1 gene in grass carp could be upregulated by bacterial infection. Overexpression of TBK1 could reduce the number of adhesive bacteria in cells and promote cellular migration, proliferation, vitality, and anti-apoptosis ability. TBK1 could activate the NF-kappa B signaling pathway and induce autophagy in cells. These findings provide important information about the defense and immune mechanisms used by grass carp against pathogens.

Summary: In this study, a recombinant baculovirus expressing VP35-VP4 proteins from GCRV was generated and used to vaccinate grass carp through oral administration. The results showed that the vaccine induced both non-specific and specific immune responses, providing protection against GCRV infection with a relative percent survival rate of 56%. This study suggests that the silkworm pupae powder could be used as an effective oral vaccine for grass carp.

Summary: In this study, a grass carp ortholog of mammalian NLK, named CiNLK2, was identified and found to be involved in the Wnt/β-catenin signaling pathway and immune response. CiNLK2 was widely expressed in various tissues and up-regulated under the treatment with poly I:C or GCRV. Overexpression of CiNLK2 suppressed IFN I production, while knockdown of CiNLK2 increased IFN I expression. CiNLK2 functioned as an inhibitor for IFN I response by targeting the MAVS-IRF3 signal axis.

Summary: This study developed a prophylactic grass carp reovirus (GCRV) vaccine using virus-like particle (VLP) assembly technology, and the highly immunogenic candidates in the vaccine were VP4, VP56, and VP35. Expression of VP4, VP56, and VP35 in both Escherichia coli and Pichia pastoris systems resulted in successful assembly of uniform, stable, and non-toxic VP4/VP56/VP35 VLPs. Vaccination with P. pastoris VLPs significantly improved survival rate, IgM levels, tissue viral load, and antioxidant immune-related indicators in grass carp.

Summary: CD3 is a crucial component of the TCR-CD3 complex in adaptive immunity, with its homologues CiCD3γ/δ and CiCD3ε identified in grass carp and shown to be highly expressed in healthy fish and modulated by infections. A monoclonal antibody against CiCD3γ/δ was successfully produced, which showed good reactivity in Western blotting and flow cytometry analysis. The CiCD3γ/δ(+) cells were found to be localized in the gills and head kidney, accounting for a small percentage of lymphocytes in different organs.

Summary: This study aimed to investigate the potential effectiveness of VD3 in treating Largemouth bass ranavirus (LMBV). The results showed that feeding Largemouth bass with 15000 IU/Kg and 20000 IU/Kg of VD3 significantly suppressed and cleared LMBV pathogens. In addition, VD3 treatment increased the expression levels of key immune-related genes and enhanced antioxidant and immune activities in the serum. Tissue pathology sections also revealed a dose-dependent relationship between VD3 supplementation and tissue damage.
