Effect of Hepatic or Renal Impairment on the Pharmacokinetics of Canagliflozin, a Sodium Glucose Co-transporter 2 Inhibitor

Purpose: Canagliflozin is a sodium-glucose cotransporter 2 inhibitor approved for the treatment of type 2 diabetes mellitus (T2DM). Because T2DM is often associated with renal or hepatic impairment, understanding the effects of these comorbid conditions on the pharmacokinetics of canagliflozin, and further assessing its safety, in these special populations is essential. Two open-label studies evaluated the pharmacokinetics, pharmacodynamics (renal study only), and safety of canagliflozin in participants with hepatic or renal impairment. Methods: Participants in the hepatic study (8 in each group) were categorized based on their Child-Pugh score (normal hepatic function, mild impairment [Child-Pugh score of 5 or 6], and moderate impairment [Child-Pugh score of 7-9]) and rceived a single oral dose of canagliflozin 300 mg. Participants in'the renal., study (8 in each group) were categorized based on their creatinine clearance (CLCR) (normal renal function [CLCR. 80 mL/min]; mild [CLCR 50 to <80 mL/min], moderate [CLcR 30 to <50 mL/min], or severe [CLCR <30 mL/min] renal impairment; and end-stage renal disease [ESRD]) and received a single oral dose of canagliflozin 200 mg; the exception was those with ESRD, who received 1 dose postdialysis and 1 dose predialysis (10 days later). Canagliflozin's pharmacokinetics and pharmacodynamics (urinary glucose excretion [UGE] and renal threshold for glucose excretion [RTG]) were assessed at predetermined time points. Findings: Mean maximum plasma concentration (C-max) and area under the plasma concentration-time curve from time zero to infinite (AUG), values differed by <11% between the group with normal hepatic function and those with mild and moderate hepatic impairment. In the renal study, the mean Cffia, values were 13%, 29%, and 29% higher and the mean AUCO3 values were 17%, 63%, and 50% higher in participants with mild, moderate, and severe renal impairment, respectively; values were similar in the ESRD group relative to the group with normal function, however. The amount of UGE declined as renal function decreased, whereas RTG was not suppressed to the same extent in the moderate to severe renal impairment groups (mean RTG, 93-97 mg/dL) compared with the mild impairment and normal function groups (mean RTG, 68-77 mg/dL). Implications: Canagliflozin's pharmacokinetics were not affected by mild or moderate hepatic impairment. Systemic exposure to canagliflozin increased in the renal impairment groups relative to participants with normal renal function.. Pharmacodynamic response to canagliflozin, measured by using UGE and RTG, declined with increasing severity of renal impairment. Summary: Al-matrix composites reinforced with varying amounts of milled carbon fibers were produced through uniball magneto milling and uniaxial hot pressing. The addition of 20% MCFs resulted in higher compressive strength and modulus of elasticity compared to other composites. Factors such as enhanced interface, porosity elimination, refined microstructure, and improved wetting contributed to the improved composite properties.

Summary: The study investigated the effects of magnetic arc oscillation (MAO) at different frequencies on the deposition of Hastelloy C276 alloy samples during wire arc additive manufacturing (WAAM). It was found that MAO could improve the surface accuracy and mechanical properties of the alloy by reducing arc density, decreasing solidification time, and enhancing dendrite growth orientations. Additionally, MAO at 5 Hz showed enhanced mechanical properties, while frequencies of 10 Hz and 20 Hz had less pronounced effects on mechanical performance improvement.

Study of wear and friction properties of a Co-free CrFeNiAl0.4Ti0.2 high entropy alloy from 600 to 950 °C

Summary: This study investigates the sliding friction and wear performance of an economical Co-free CrFeNiAl0.4Ti0.2 high entropy alloy over a wide temperature range. The results show that sliding temperature has a significant influence on the friction coefficient, wear rate, and worn surface morphology. The maximum friction coefficient and minimum wear rate are achieved at 800 degrees C. The main wear mechanism is abrasion at 600 degrees C, but oxidation becomes more significant at higher temperatures.

Summary: In this study, the microstructure and mechanical properties of wire arc additively manufactured Ti6Al4V alloy with/ without magnetic arc oscillation were investigated. The results showed that the addition of magnetic arc oscillation resulted in refinement of the microstructure, modification of texture orientation, and improvement of mechanical properties of the deposition.

The Effect of Strain in Recrystallization Region on Interphase Precipitation and Mechanical Properties of Ferritic Microalloyed Steels

Summary: High yield strength ferritic microalloyed steels with the addition of Nb, Mo, V, and Cr microalloying elements and controlled thermomechanical treatment have been developed. The microstructures and mechanical properties are characterized using advanced microscopy techniques and uniaxial tensile testing. By adjusting the roughing strain in the recrystallization region, the grain size of the steel can be reduced, and the number density and size of NbV carbonitrides and VCr interphase precipitates can be increased. These finer grains and more nanosized interphase precipitates are key to achieving high yield strength.

Tribological performance of a cost-effective CrFeNiAl0.3Ti0.3 high entropy alloy based self-lubricating composite in a wide temperature range

Summary: A cost-effective self-lubricating composite material based on CrFeNiAl0.3Ti0.3 as the matrix and Ag as the solid lubricant has been fabricated. Tribological tests show that the presence of Ag tribo-film reduces the coefficient of friction and wear of the composites from room temperature to 600 degrees C. The wear mechanism changes from abrasive to oxidative wear at 750 degrees C.
