Disposal of iron by a mutant form of lipocalin 2

Iron overload damages many organs. Unfortunately, therapeutic iron chelators also have undesired toxicity and may deliver iron to microbes. Here we show that a mutant form (K3Cys) of endogenous lipocalin 2 (LCN2) is filtered by the kidney but can bypass sites of megalin-dependent recapture, resulting in urinary excretion. Because K3Cys maintains recognition of its cognate ligand, the iron siderophore enterochelin, this protein can capture and transport iron even in the acidic conditions of urine. Mutant LCN2 strips iron from transferrin and citrate, and delivers it into the urine. In addition, it removes iron from iron overloaded mice, including models of acquired (iron-dextran or stored red blood cells) and primary (Hfe (-) (/) (-)) iron overload. In each case, the mutants reduce redox activity typical of non-transferrin-bound iron. In summary, we present a non-toxic strategy for iron chelation and urinary elimination, based on manipulating an endogenous protein: siderophore: iron clearance pathway.

Flavonoids in Lu'an GuaPian tea as potential inhibitors of TMA-lyase in acute myocardial infarction

Summary: Current studies have found that plasma trimethylamine N-oxide (TMAO) level is positively correlated with the risk of acute myocardial infarction (AMI). Flavonoids in Lu'an GuaPian tea, especially kaempferol 3-O-rutinoside, have shown potential in inhibiting TMA production and could be used for AMI treatment.

Chemical composition and antibacterial activity of 12 medicinal plant ethyl acetate extracts using LC-MS feature-based molecular networking

Summary: The study evaluated the antibacterial activities of 12 medicinal plant extracts against six foodborne bacteria, with Toona sinensis shoots and Firmiana simplex barks showing the highest inhibitory activities against various strains. Firmiana simplex barks exhibited strong antibacterial effects against Salmonella typhi. Furthermore, the identified bioactive compounds in the plant extracts were mainly polyphenols, glycosides, and terpenoids.

Discovery of Camellia sinensis catechins as SARS-CoV-2 3CL protease inhibitors through molecular docking, intra and extra cellular assays

Two novel enantiomers from metarhizium flavoviride and their inhibitory activities against α-glucosidase

Summary: In this study, a pair of novel enantiomers, metarrhiziumoids A and B, were isolated from the extract of Metarhizium flavoviride. The structures of the compounds were identified using various techniques. The compounds showed glycosidase inhibitory activity, with metarrhiziumoids B exhibiting stronger activity than metarrhiziumoids A. Molecular docking results revealed different binding energies between the two enantiomers, which may contribute to the differences in inhibitory activity.

Improving flavor of summer Keemun black tea by solid-state fermentation using Cordyceps militaris revealed by LC/MS-based metabolomics and GC/ MS analysis

Summary: This study aimed to improve the flavor of summer Keemun black tea (KBT) using Cordyceps militaris solid-state fermentation. The analysis of volatile and non-volatile components revealed that the total polyphenol, total flavonoid, and total free amino acid contents were reduced after solid-state fermentation. Additionally, the contents of non-galloylated catechins and D-mannitol increased, while the galloylated catechins and flavonoid glycosides decreased, leading to a decrease in bitterness and astringency of KBT. Aroma compounds such as dihydro-beta-ionone and beta-ionone were identified as the key contributors to the woody and floral odors in the fermented KBT.

Cyclic pentapeptides with anti-inflammatory, cytotoxic or ?-glucosidase inhibitory activities from Basidiobolus meristosporus

Summary: Nine undescribed cyclic pentapeptides, named basidiosin D-L, were isolated from mycelia of B. meristosporus RCEF4516 using semi-preparative HPLC. These compounds showed inhibitory effects on NO production in LPS-activated RAW264.7 cells and cytotoxicity against RAW264.7, 293 T, and HepG2 cells. Additionally, they exhibited stronger inhibitory effects on alpha-glucosidase compared to acarbose, except for compound 7.

Virtual communication is commonly used in Finnish interstitial lung disease multidisciplinary meetings

Summary: This study investigated the implementation and key elements of Interstitial Lung Diseases (ILD) Multidisciplinary Meetings (MDMs) in Finnish specialized care. ILD MDMs were organized approximately every two weeks in all academic centers of Finland, with key participants including a radiologist, respiratory physicians, junior staff, pathologist, and a rheumatologist. Virtual communication was commonly used in ILD MDM implementation, particularly in small centers located in Eastern and Northern Finland. Virtual access to ILD MDMs of academic centers was available in most parts of Finland, enabling small centers to benefit from their expertise.

Novel methylated flavoalkaloids from Echa 1 green tea inhibit fat accumulation and enhance stress resistance in Caenorhabditis elegans

Cyclic Peptides from the Opportunistic Pathogen Basidiobolus meristosporus

Summary: Three new cyclic peptides, meristosporins A, B, and C (1-3), with unique amino acid composition, were isolated from Basidiobolus meristosporus, an opportunistic pathogen. The structures of these compounds were determined using advanced analytical techniques. The biosynthetic pathway of meristosporin A (1) was inferred based on genetic analysis. Compounds 1 and 2 exhibited cytotoxicity towards RAW264.7 and 293T cells, respectively, suggesting their potential involvement in fungal-induced cell injury.

Two new catechins from Zijuan green tea enhance the fitness and lifespan of Caenorhabditis elegans via insulin-like signaling pathways
