Autophagy in T-cell differentiation, survival and memory

Liqing Wang, Jugal Kishore Das, Anil Kumar, Hao-Yun Peng, Yijie Ren, Xiaofang Xiong, Jin-Ming Yang, Jianxun Song

Summary: Autophagy has emerged as a critical regulatory mechanism of the immune system, playing crucial roles in T cell biology and the fate of different T cell subsets. It maintains T cell development and survival by regulating the degradation of organelles and apoptotic proteins, and impacts the formation of memory T cells. Alteration of autophagy in T cells may lead to pathological conditions such as inflammation, autoimmune diseases, and cancer.

Stem Cell-Derived Viral Antigen-Specific T Cells Suppress HIV Replication and PD-1 Expression on CD4+T Cells

Summary: Antigen-specific CD8+ cytotoxic T lymphocytes derived from induced pluripotent stem cells (iPSCs) can efficiently suppress HIV replication and protect CD4+ T cells. Adoptive transfer of iPSC-CTLs significantly inhibited HIV replication in mice and promoted the generation of persistent anti-HIV memory T cells.

T Cell Response to SARS-CoV-2 Coinfection and Comorbidities

Liqing Wang, Hao-Yun Peng, Aspen Pham, Eber Villazana, Darby J. Ballard, Jugal Kishore Das, Anil Kumar, Xiaofang Xiong, Jianxun Song

Summary: Over the past three years, COVID-19 has emerged as a growing global health concern. We have summarized the performance of T cells during coinfection of SARS-CoV-2 with other viruses, bacteria, and parasites, and determined if these alterations in T cell status affect clinical outcomes. Additionally, we have investigated T cell alterations in patients with diabetes, asthma, and hypertension during SARS-CoV-2 infection and summarized whether changes in T cell response influence clinical outcomes in comorbidities.

Identification of a NACC1-Regulated Gene Signature Implicated in the Features of Triple-Negative Breast Cancer

Chrispus M. Ngule, Hami Hemati, Xingcong Ren, Oluwafunminiyi Obaleye, Amos O. Akinyemi, Felix F. Oyelami, Xiaofang Xiong, Jianxun Song, Xia Liu, Jin-Ming Yang

Summary: In this study, stemness-associated genes involved in the progression of triple-negative breast cancer (TNBC) were identified using bioinformatics approaches. These genes were found to be correlated with tumor hypoxia, stemness enrichment, and immune-suppressive tumor microenvironment.

Control of CD4+ T cells to restrain inflammatory diseases via eukaryotic elongation factor 2 kinase

Hao-Yun Peng, Liqing Wang, Jugal Kishore Das, Anil Kumar, Darby J. Ballard, Yijie Ren, Xiaofang Xiong, Paul de Figueiredo, Jin-Ming Yang, Jianxun Song

Summary: The deficiency of eEF2K disrupts the survival and function of CD4(+) T cells, leading to excessive production of inflammatory cytokines and increased metabolic activity and mitochondrial bioenergetics. The absence of eEF2K exacerbates the severity of inflammation-related diseases, but targeting eEF2K with drugs can alleviate symptoms.

An optimized protocol for the generation of HBV viral antigen-specific T lymphocytes from pluripotent stem cells

Mohammad Haque, Xiaofang Xiong, Fengyang Lei, Jugal Kishore Das, Jianxun Song

Summary: This study demonstrates the generation of HBV Ag-specific CTLs from induced pluripotent stem cells for cancer therapy, showing promising results in suppressing HBV replication in a murine model. The approach of using iPSC-derived CTLs may hold potential for chronic viral infections.
